Objective The purpose of this study was to identify Hsf1as a target gene of AR inSertoli cell and its molecular mechanism in male infertility.Materials and Methods PCR and Western blot were used to comparing theexpression level of heat shock transcription factor-1(Hsf1) in Sertoli cell-selectiveknockout AR mice (S-AR-/y) with wild type mice (WT) and TM4cell line with orwithout testosterone (T). luciferase assay, electrophoretic mobility shift assay (EMSA)and chromatin immunoprecipitation assay (ChIP) were used to identify AR directlyregulated Hsf1expression via DNA-binding specific promoter.Results Hsf1expression level in S-AR-/ymice was significantly increased comparedwith WT mice. On condition of T, Hsf1expression of TM4cell line were significantlyreduced both mRNA and protein level. And we revealed that AR directly regulated Hsf1expression via DNA-binding specific promoter. Moreover, our data suggests that HSF1acts as a positive factor for target gene expression, in particular regulating heat shockprotein105(HSP105) and HSP60.Conclusion These results suggest that the novel roles of the AR-regulated HSF1andpossibly HSPs in the control of spermatogenesis.
