Objective:Study the osteoconduction and tissure biocompatibility of Demineralized Dental Matrixes(DDM) made by various methods in vivo by ectopic osteogenesis test, to prepare the basic datas for exploring into osteoconduction mechanism of the DDM.Method:Collect extracted teeth by a unified standard,and Demineralized Dental Matrixes(DDM) made by two regular ways of foreign, and DDM muscle ectopic implantation modelling in buttock muscle of4Beagle dogs.According to the implanted materials,five groups will be coded as A,B,C,D,E,and DMM1in A group, DMM1in B group(DMM1by Inactivation treatment), DMM2in C group, DMM2’in D group(DMM2by Inactivation treatment),DMM3in E group(commercial). A and B group will be in buttock muscle site, C,D,E group in right buttock muscle, four periods set for each site. Samples taken in2W,4W,6W,8W postoperatively. the alkaline phosphatase activity was measured and a gross histologic analysis was carried out. SPSS17.0used for data analysis.Result:1.Gross observation showed:diets and activities of all Beagle dogs were normal after operation, the wound healed well in a week, no congestion and edema were observed.2.Microstructure observation:During the whole observation period, materials were elosely integrated with the surrounding musele tissue, and the interface between muscle and material revealed no signifieant fiber wall formation. no edema, infection and necrosis were observed.3.Histological Observation:ectopic osteogenesis was observed in commercial group in8weeks, osteoid formation was observed, in which trabecular bone was found. No bone formation was found in other groups.4.ALP Examination:statistical difference was shown when comparing ALP of commercial DDM in different time spots(P<0.01), there is no statistical difference in the rest of the groups in their chosen time. The ALP of commercial DDM was higher than other groups, there is statistical difference in their chosen time.(P<0.01)Conclusion:All DDM showed good biocompatibility in chosen time in this experiment, but bone induction activity of self-made DDM was not present. Good bicompatibility and bone induction activity of DDM3(commercial DDM) was observed.
