Objective:To screen and identify serum microRNA(miRNA) as promising biomarkers in hereditary nonpolyposis colorectal cancer(HNPCC) by using microRNA arry and qRT-PCR(real-time quantitative PCR) for early screening and diagnosis.Methods:1.Samle collection:(1)Collecting Amsterdam II clinical standards of HNPCC family members’serum samples, combined with clinical data, sample source, family characteristics and other information to integrated assessment.(2)Collecting normal persons who’s families have no history of HNPCC as contol group.2.Screening serum miRNA as promising biomarker in hereditary nonpolyposis colorectal cancer by using microRNA arry.3.Using3target gene prediction softwares(mirnada,targetscan,mirbase) to predict the serum microRNAs from the array,and check out the microRNAs who has target genes by qPCR.Results:1.Collecting6HNPCC families samples,including10HNPCC patients,9normal persons from HNPCC families and6normal persons without HNPCC history, all the samples are serum.2.We found57up-regulated miRNAs and30down-regulated miRNAs by miRNA array and chosen7miRNAs which are more differential expression in patients serum.3.Utilizing3target gene prediction softwares,taking intersection of the3softwares’ results that targets belonged to miR-20a-5p,miR-548b-5p and miR-548as-3p.Then using qPCR to check out,and we found that serum miR-548as-3p’s result corresponded with the miRNA array’s result:its differential expression in hereditary nonpolyposis colorectal cancer is significant.But the result of qPCR from miR-20a-5p and mir-548b-5p are not corresponded with the miRNA array.Conclusion:Our study domenstated that there were differential expressions in serum microRNAs in NPCC patients,and that miR-548as-3p could potentially serve as biomarkers for this desease.
