Eucommia cortex is a famous tonic traditional Chinese drug. In recent years, many researchers from abroad and China have made much efferts to study on the Eucommia cortex. They have found that Eucommia cortex can strengthen the liver and kidneys, strong bones, tocolysis, lower blood pressure and anti-fatigue, so it is used for treating liver and kidney deficiency, weak waist, impotence, hypertension, hemorrhage of pregnancy and so on. Besides that it has been demonstrated that Eucommia cortex could be used for the osteoporosis treatment. But the studies of Eucommia cortex on osteoporosis have mostly concerned about that the differentiation and proliferation of osteoblast by Eucommia cortex and metabolism in the rats. So the mechanism of Eucommia cortex on the treating osteoporosis has not been well understood. This research concerns about the mechanism of Eucommia cortex on treating osteoporosis, especially studing the mechanism of Eucommia cortex on the OPG/RANKL ratio of MC3T3-E1osteoblasts. This research shows a new mechanism of Eucommia cortex on treating osteoporosis via OPG/RANKL/RANK system.This research was saperated two parts. The first part concerned about the proliferation and differentiation of MC3T3-E1osteoblasts via Eucommia cortex. The second part was about the ratio of OPG/RANKL by Eucommia cortex.In the first part, the MTT was used to found the fitable concerntration of Eucommia cortex on MC3T3-E1osteoblasts. Then the MC3T3-E1osteoblasts were cultivated by high, medium and low concerntration of Eucommia cortex. The MTT was used again to test the proliferation of MC3T3-E1osteoblasts by different concerntration of Eucommia cortex. The ALP was used to test the differentiation of MC3T3-E1osteoblasts by different concerntration of Eucommia cortex. The Alizarin red S was used to see the mineralization of MC3T3-E1osteoblasts by Eucommia cortex also to test the differentiation of the MC3T3-E1.In the second part, the RT-PCR was used to test the transcription of OPG and RANKL of MC3T3-E1osteoblasts by Eucommia cortex firstly. Then the Elisa kit was also used to test OPG and RANKL of MC3T3-E1osteoblasts by Eucommia cortex and calculated the ratio of OPG/RANKL.It was found that the Eucommia cortex could increase the proliferation and differentiation of MC3T3-E1osteoblasts and also increas the expressing the OPG, but inhibiting the expressing the RANKL of MC3T3-E1osteoblasts. What’s more, the ratio of OPG/RANKKL of MC3T3-E1osteoblasts was up-regulated by Eucommia cortex. According to the results, it was shown that Eucommia cortex could be benefit for the bone remolding by increasing the osteoblasts and inhibiting osteoclasts by up-regulating OPG/RANKL ratio of osteoblasts. Therefore the Eucommia cortex was benefit for the therapy on osteoporosis.
