Objective:To explore cilostazol and losartan effects on transient outward potassium current and L-Type calcium current in isolated rat right ventricular myocytes. To explain their possible anti-arrhythmic mechanism.Method:Male SD rats was randomly divided into three groups:control group, cilostazol group and losartan group. Cilostazol group and losartan group rats were given10mg/kg/d cilostazol and10mg/kg/d losartan by oral administration for2-3weeks, respectively. Then single myocytes were isolated from the right ventricles of SD rats. Experimental conditions were set as(1)control group:Ito ionic current was recorded after5min of equilibration with normal extracellular solution and2min of equilibration with NMDG extracellular solution for control data, and then recorded after add cilostazol of50μmol/L or2μmol/L.(2) cilostazol group and losartan group: Ito ionic current was recorded after5min of equilibration with normal extracellular solution and then2min of equilibration with NMDG extracellular solution.(3) Ica-L ionic current of three groups were recorded after5min of equilibration with normal extracellular solution.Results:Cilostazol of50μmol/L and2μmol/L significantly decreased Ito. The peak amplitude of decreased from (21.55±2.61)pA/pF to (5.36±2.16)pA/pF, and (18.64±7.89)pA/pF to (7.63±1.78)pA/pF, respectively。There are no difference to compare with Ito and Ica-L in control group and cilostazol group, the same to losartan group.Conclusion:The presenr study demonstrated that cilostazol inhibited Ito. It is suggest that cilostazol may has effect to ventrieular arrhythmia.
