The Expression Of FOXP3and Its Prognosis In Diffuse Large B Cell Lymphoma

ObjectiveTo analyze the expression of FOXP3in diffuse large B cell lymphoma (DLBCL) tissues and its subtypes and further explore the significance of FOXP3+cells in tumor cells and peritumoral tissues.MethodsCD20and FOXP3were assessed by immunohistochemistry and immunofluorescence double staining in164cases of diffuse large B cell lymphoma. We analyzed the clinical and pathologic data of all the cases in retrospective study. Then, we detected the expression of FOXP3in different subtype (include GCB and non-GCB) of DLBCL cell lines by PCR and Western Blot.Results1The expression of FOXP3and CD20were observed within tumor bed and in the tissue surrounding tumor through the immunohistochemical staining. The expression of FOXP3and CD20presented in diffuse pattern and those in tumor cells were more abundant and intended than in tissue surrounding tumors.2High density of FOXP3in the tumors had relationship with later stage and invoved extranodal involvement (P<0.05), decreased overall survival (OS). Furthermore, High density of FOXP3in tissue surrounding tumors was significant positive correlated with later LDH (P<0.05).3Cox regression analysis showed that the expression of IPI and FOXP3was independent prognosis factor of DLBCL (P<0.05).4After RT-PCR and western blot, we found the expression of FOXP3both expressed in non-GCB cell line(LY-3) and GCB cell line(LY-7).ConclusionThe prognostic significance of FOXP3in diffuse large B-cell lymphoma depends on the the FOXP3distribution and density. The density of Intratumoral FOXP3was an independent predictor of overall survival in diffuse large B-cell lymphoma. Moreover, the density of Intratumoral FOXP3was closely correlated with extranodal involvement and poor prognosis. This may provide new treatment for different subtype of diffuse large B cell lymphoma.
