Objectives:this study uses Thymus tolerance to develop animmune system that is tolerant to MBP/Autologous brain cells/Autologous Cerebrospinal fluid so that autoimmune injury can be suppressed following surgical disruption of the blood-brain barrier.Methods:Part1,a study of injecting MBP or Autologous brain cells into the Thymus tissues.Injecting MBP or Autologous brain cells and or PBS buffer solution to the Thymus tissues after the the rabbit models of SBI were taken. All rabbits were divided into6groups. Groups A (PBS)/B(Brian Cells)/C(MBP) were drawn blood on1st day,3rd day,7th day,14th day to measure levels of IL-1, IL-2, IL-10, TGF-β by ELISA test. All the rabbits of A/B/C were killed14days postoperatively to measure the levels of Foxp3or Bcl-2of the rabbit Thymus tissues through immunofluorescence histochemistry. The rabbits of Groups D(PBS)/E(Brian Cells)/F (MBP)were killed7days postoperatively to measure the levels of Fas/FasL of the brain tissues;Part2,a study of injecting Autologous Cerebrospinal fluid into the Thymus tissues. Injecting Autologous Cerebrospinal fluid or PBS buffer solution to the Thymus tissues after the the rabbit models of SBI were taken. All the rabbits were drawn blood on1st day,3rd day,7th day,14th day to measure levels of IL-1, TGF-β by ELISA test.Injecting Autologous Cerebrospinal fluid to the Thymus tissues demonstrated decreased levels of IL-1and increased levels ofTGF-β following SBI.Results:Injecting MBP or Autologous brain cells or CSF into the Thymus tissues demonstrated decreased levels of IL-1, IL-2, BCL-2and increased levels of Fas/FasL, Foxp3, IL-10, TGF-β following SBI.Conclusions:Developing thymus immunological tolerance through thymus injection of MBP/brain cells/CSF can reduce autoimmune injury following the SBI and contribute to the recovery of the immune privilege to the central nervous system because of the time-limited duration of tolerance following the rehabilitation of the blood-brain barrier.
