Objective:To Observe the expression of caspase-8, NF-kB in the endometriosis after treatment of Genistein, to affect the relationship between the expression of Caspase-8and NF-kB treated rat endometriosis in Genistein.Methods:①Establishment of rat endometriosis model.The6-8weeks old female SD rats were selected and subcutaneous injected diethylstibestrol to achieve the same estrous cycle, transpcanted the autologous endometrial in the abdominal wall fascial layer, then injected17-beta-estradiol dissolved in0.1ml sesame oil every three days, a total of3weeks.Treatment method and does selection, testing graft volume.②The SD rats were randomly divided into three groups, Group Ⅰ (model group, n=15), GroupⅡ (lower-dose Genistein,0.5mg/kg/d, n=15), GroupⅢ (high-dose Genistein,50mg/kg/d, n=15).Group I subcutaneous injected sesame oil0.05ml/(kg.d) once daily; Group Ⅱ、GroupⅢ were given Gen0.5mg/kg/d、50mg/kg/d, dissolved in0.1ml sesame oil once daily subcutaneous injection, a total of12weeks.③Histological examination, using HE staining to observe the pathological changes of endometrial tissue transplantation.④We performed SP-9000to detect tissue specimens of Caspase-8, NF-kB expression.Results:①Change of graft volume:As Gen does increased, the eutopic endometrium volume tends to smaller.Compared with the control group, Gen low dose group of graft volume did not change significantly, no significant difference between two groups (P>0.05).Spatially in the high does group it was siginificantly decreased (P<0.01).②Pathological changes:compared with the control group, Gen treatment group transplanted glands atrophy, the visible portion of shorter, disappear, the epithelial cell layer thickness, visible part of glandular epithelial shedding, unclear structure. The Gen effect of high dose group was the most obvious.③Campared with the model group, the expression rate of Caspase-8was significantly increased in the treatment groups (P<0.05); the positive rate of Caspase-8protein expression was80%in the GroupⅢ(high-does Genistein), higher than69.23%in the Group Ⅱ (lower-does Genistein)(P<0.05)。④Compared with the control group, Gen low dose group and high dose group in the expression of NF-kB was no significant difference (P>0.05)Conclusion:1.In rats endometriosis treated with Genistein, the expression of Caspase-8was increased, showed that Caspase-8regulates apoptosis, participates in the Gen for the treatment of endometriosis occurrence, development process.2.Gen has a good inhibiting the hyperplasia of rat EMs, promote cell apoptosis. But its mechanism of action may not be through the NF-kB pathway, but of its effect through other pathways.
