Objective:In this paper, the effect of betulin on ovarian cancer CAOV3cells and the mechanism of apoptosis-induced were investigated. A series of studies has been made for seeking out better nature substances for applying to clinic.Method:MTT assay was used to investigate the effect of betulin on cultured CAOV3cells and the IC50value was determined. To assess whether betulin induces apoptotic cell death in CAOV3cells, morphological analysis and flow cytometric analysis were employed; in vitro Caspase-3,-9activity and PARP cleavage were analyzed in betulin treated CAOV3cells.Result:1. Betulin obviously suppressed the survival of ovarian cancer CAOV3cells, and inhibitive effect increased with the concentration of betulin. IC50value was8μg/ml.2. Betulin (10μg/ml) induce apoptotic cell death, as evidenced by morphological characteristics such as membrane blebbing, nuclear condensation and fragmentation, and apoptotic body formation.3. Flow cytometric analysis showed that the total rate of apoptosis cells was reached62.77%at12h after betulin treatment, in contrast, that of control cells were only1.45%.4. The immunobloting analysis showed that PARP, a substrate of Caspase-3, cleaved to yield the85KDa version of this protein after12h treatment of betulin in CAOV3cells.5. In vitro Caspase activity analysis showed that both Caspase-9and Caspase-3was activated in betulin treated CAOV3cells.Conclusion:Betulin induces Caspase-3、 Caspase-9associated apoptosis in ovarian cancer CAOV3cells.
