Objective: Understand the clinical features of malignant melanoma in Xinjiang region,ascertain C-KIT gene mutations and the CD117expression of malignant melanoma inXinjiang region. Analyzed the relationship between C-KIT gene mutations and clinicalphenotypes of malignant melanoma. So to discuss its relationship with the occurrence,development and prognosis of malignant melanoma, in order to find a theory basis formolecular target therapy of malignant melanoma in Xinjiang area.Methods: Collected theXinjiang uygur autonomous region people’s hospital confirmed the clinicopathologicaldata of76patients with malignant melanoma from2005to2011. Using specific primerpolymerase chain reaction PCR and DNA direct sequencing method to detect76cases ofmalignant melanoma and40cases of normal paraffin embedding of skin tissue C-KITgene mutation of exons11and13.Using immunohistochemical method to detect76casesof malignant melanoma and30cases of pigmented nevus paraffin embedding tissue ofCD117expression. The relationship between C-KIT mutation and clinical phenotype wasanalyzed by2test or Fisher’s exact test according to the SPSS17.0Software, statisticalsignificance was considered when P <0.05.Results:1. the average age of Patients is62.2years old, under the age of60group C-KIT gene mutation rate is increased more than60years old age group.(2=6.613, P <0.05)2.12of the76patients of malignantmelanoma were detected the C-KIT gene mutation, Mutation rate is15.79%(12/76);There are10mutations in exon11, All are point mutations, mutation rate is83.33%(10/12); Another2cases of point mutations are in exon13, mutation rate is16.67%(2/12);40cases of normal skin tissue has not been found that the gene mutation.3.69cases of76patients with malignant melanoma were detected positive CD117expression in thisgroup,9cases were detected negative expression;18cases of30patients with pigmentednevus were detected positive CD117expression in this group,12cases were detectednegative expression. The positive CD117expression rate in malignant melanoma aresignificantly more than pigmented nevus. The difference has statistically significant(2=5.83, P <0.05).4. The C–KIT gene mutations rate in mucous membranes, acral, non-acral malignant melanoma tissue were18.2%(4/21),14.7%(5/34) and41.7%(10/24),The difference has statistically significant(2=6.167, P <0.05).5. The mutations ofC-KIT in malignant melanoma patients were no significant correlation with gender,ethnicity, metastasis of lymph node(P>0.05).Conclusion:1.malignant melanoma inXinjiang region mainly occurs in the elderly, with small gender differences,acral ispredilection sites.2.C-KIT gene is one of the hot spot mutations in malignant melanomaof Xinjiang region, and mainly in with exon of L576P mutation, in order to find a theorybasis for molecular target therapy of malignant melanoma in xinjiang area.3.The C-KITmutations between Han population and Uyghur population were no difference, Initialprompt is that the C-KIT mutations may not be ethnic differences in melanoma.4.The C-KIT gene mutation of melanoma patients is closely related to the onset age, disease area,but no obvious correlation with gender, metastasis of lymph node.5. CD117expressionincreased significantly in malignant melanoma, but no obvious correlation with Onset age,gender and onset of parts. |
