Objective:The human papillomavirus16(HPV16) is an important pathogenic factorof Kazakh’s esophageal squamous cell carcinoma. The abnormal expression ofmetalloproteinase and its inhibitor are correlated with the invasion and metastasis oftumor. Here we initially discuss the correlation between the pathogenic factor of HPV16,MMPs of esophageal squamous cell carcinoma and its inhibitor TIMPs. Methods:1.RT-PCR was applied to detect the expression of MMP-2, MMP-9, TIMP-1andTIMP-2mRNA in38cases Kazakh’s esophageal squamous cell carcinoma tissues,analysis of both in cancer and distal normal tissue and the relationship between cancertissue and clinical indicators.2. Constructing the HPV16E6, E6E7fusion gene eukaryoticexpression vector, transfected to the esophageal the cell lines KYSE450by transienttransfection technique detecting the expression changes of MMP-2, MMP-9and TIMP-1and TIMP-2mRNA. Results:1. In38cases of esophageal squamous cell carcinoma anddistal normal esophageal tissue, HPV16positive expression rate was68.4%higher than ofthe distal normal esophageal tissue44.4%, the difference was statistically significant(P<0.05);MMP-9, TIMP-1and TIMP-2mRNA expression level in cancer tissue weresignificantly higher than remote normal tissues, statistically significant (P<0.05).2: Therewas no correlation at the transcriptional level of HPV16and MMP2, MMP-9andTIMP-2in cancer and distal normal tissue (P>0.05), and TIMP-1showed a positivecorrelation (r=0.424, P=0.006); There has no any correlation between indicators ofclinical pathology analytical results of HPV16and MMP2, MMP-9, TIMP-2, but it hasa positive correlation withTIMP-1in poorly differentiated (r=0.612, P=0.014) andlymph node metastasis (r=0.586, P=0.012).3: Results of cell transfection showed thatexpression level of MMP-9mRNA in E6, E6E7vector transfection group wassignificantly lower than that of the normal group and empty vector transfected group, thedifference was statistically significant (P<0.05), TIMP-1mRNA in E6E7vectortransfection group was significantly higher than the other groups, the difference was statistically significant (P<0.05). Conclusions: In the development process of Kazakh’sesophageal squamous cell carcinoma there is a certain correlation between HPV16andTIMP-1at the transcriptional level. E6, E7genes in KYSE450cell promote TIMP-1expression at the transcriptional level, while they inhibit the expression of MMP-9. Therehave been certain regulatory relationship between HPV16E6, E7genes and invasion,metastasis of esophageal cancer...
