Uncaria sessilifructus Roxb.(USR.), Dai people in Xishuangbanna named“Huai Mianwang”, is a liana in the family Rubiaceae. It mainly distributed in Yunnan,Guangxi and Guangdong province of China. USR. is the conventional medicine usedby doctor in Xishuangbanna dai hospital and the prescription medicine ofXishuangbanna dai hospital. Widespread used in Xishuangbanna. The plant isconsidered has the effect of clear away heat and toxic materials, relieve swelling, easepai and expelling wind to get through blood. The Dai people used USR.’s hooks andramulus to cure injuries from falls, rheumatoid arthritis, giddily, cold and scare,teched, bellyache, hysteritis, hemiplegia, sciatica, high blood pressure, ulcer etc..It hasa very importent medicinal value. But from now on, about the chemical compositionsof the medicinal material research reports only before one review by Phillipson, J.D in1978about Uncaria genus cover15pentacyclical indoles alkaloids and a report aboutessential oil in2012, there is not research about the non-alkaloid.Objective:Analysis of chemical compositions in Dai medicine USR., determine itsefficacy material, in order to procide scientific support for its ethnomedical uses.Methods:(1)To study the chemical constituents from95%ethanol extract ofUSR. by extensive column chromatography method;these compands were identifiedby various sepectroscopic analysis.(2) Screening test model for macrophages in mice,use Griess reagent chromogenic method to testing generation of nitric oxide. With thenumber of cells use MTT assay to detect survival cells and use the Reed and Muenchmethod calculated these compounds’ IC50values. Results:Thirty compounds were isolated from95%ethanol extract of USR. andtwenty-six of those compounds were identified by various sepectroscopic analysis.Compared with the literature data their structures were identified as23non-alkaloids:secouncarilic acid (1), uncarilic acid (2), quinovic acid (3), ursolic acid (4),3-O-[β-D-Quinovpyanosyl] quinovic acid (5),3-O-[β-D-glucopyranosyl]-quinovicacid (6),3β,6β,19-trihydroxy-23-oxo-urs-28-oic acid (7),3β,6β,19-trihydroxyurs-12-en-28-oic acid (8),3β,6β,19,23-tetrahydroxyurs-12-en-28-oic acid (9), friedelin(10), betulin (11), obtusalin (12), stigmast-4-en-3-one (13), β-sitosterol (14),β-daucosterol (15),24-en-cycloartenone (16), squalene (17), vitaminE (18),-tocopherolquinone (19),(-)-alloaromadendrene (20), eugenin (21), scopoletin (22),bis(2-ethylhexyl)phthalate (23) and3alkaloids: mitraphylline (24), isomitraphylline(25), isomitraphylline N-oxide (26). Compounds (1-2) are two new oleanane-typetriterpenoid,(1-23) were isolated from USR. For the first time and compounds (5,6,9,13,16,19) were reported from Uncaria for the first time. Compounds24-26wereindole-type alkaloid are the main active substance in U. sessilifructus Roxb. Thecompounds11,12were lupane-type triterpenoid, the compound16was cycloartane-type triterpenoid, the compound10was feiedelane-type triterpenoid and compounds3-9was ursane-type triterpenoid. Both of compounds1-2,3-9,24-26showedno-observed activity inhibit NO production.Conclusion:Triterpene, steroid(glucoside) and indole alkaloid are the mineeffective chemical constituents containd by USR. Twenty-six compounds wereisolated from USR. All these compounds were found in USR. for the first time exceptalkaloids. These compounds’ finding provides the basis for scientific for furtherresearch on the dai medicine USR.’s Pharmaceutical development.
