Objective: To investigate the effects of simvastatin on the apoptosis of humanumbilical vein endothelial cells induced by sepsis rats serum and to explore its potentialmolecular mechanism.Methods: Using the modified cecal ligation and puncture replication model ofsepsis, collected serum standby. In vitro culture of human umbilical vein endothelial cell(ECV-304) were divided into4groups (control group, sepsis group, simvastatinintervention group, simvastatin group).Culture of24h.Mtt colorimetry were performed toconfirm cell apoptotic ratio; Acridine orange staining was used to observe the renal cellapoptosis morphologic changes;Reverse transcriptase-polymerase chain reaction was usedto detect the quantity of mRNA Bcl-2and Bax;Cell apoptosis was detected by flowcytometry rate.Results:1、MTT: Compared with the sepsis group, simvastatin intervention group increased theactivity of cells.2、AO staining: The control group and simvastatin cell morphology were normal; sepsisgroup showed large amounts of apoptotic cells; and simvastatin intervention group withsmall quantities of apoptosis.3、 RT-PCR: Compared with the control group, the sepsis group, simvastatinintervention group, The mRNA of Bcl-2and Bax expression levels wereup-regulated(P<0.01). Compared with the sepsis group, simvastatin intervention group themRNA of Bcl-2expression levels were up-regulated (P<0.05), while the mRNA of Baxexpression levels were down-regulated (P<0.01).4、Flow cytometry:Control group cell apoptosis rate of4%, sepsis septic group cellapoptosis rate of30%, simvastatin intervention group the apoptosis rate of17%. Conclusions:Simvastatin can reduce the number of apoptosis of human umbilical vein endothelialcells induced by sepsis rats serum, the underlying mechanism might be related to upregulation of Bcl-2and down regulation of Bax.
