Objective:The study was performed to investigate the influence of biological behaviour ofAnnexin A1in Nasopharyngeal Carcinoma (NPC) cells, and initially explore theeffect of Annexin A1in NPC invasion and metastasis, and analyze its potentialmechanisms.Methods:NPC cell lines5-8F with higher transfer and6-10B with lower transfer wereselected and the level of Annexin A1protein in both cells were detected by Westernblot. Recombinant plasmids pEGFP-C1-Annexin A1andpRNAT-U6.1-siRNA-Annexin A1were constructed, and transfected into5-8F and6-10B cells by liposome-mediated transfection, respectively, then total protein wasprepared and the level of Annexin A1was assessed by western blot.,the stable celllines of the5-8F/pEGFP-C1-Annexin A1and6-10B/pRNAT-U6.1-siRNA-AnnexinA1were screened, the change of Annexin A1in two groups cells phenotype wereobserved by experiments of CCK-8, flow cytometry, soft agar colony formation, andTranswell migration and invasion.Results:(1) The level of Annexin A1was lower in5-8F cells and higher in6-10B cells.(2) The up-regulation levels of Annexin A1expression inhibited the growth ofNPC cells, enhanced the blocking ability of G0/G1cycle, weakened the abilityof parking non-dependent growth and the ability of migration and invasion.conversely, The down-regulation levels of Annexin A1expression promotedthe growth of NPC cells, reduced the inhibition ability of G0/G1cycle,reinforce its ability of parking non-dependent growth and the ability ofmigration and invasion.Conclusions:(1) Recombinant expression vector pEGFP-C1-Annexin A1and interferencevector pRNAT-U6.1-siRNA-Annexin A1were successfully constructed. (2) Up-regulation the level of Annexin A1suppresses the growth, proliferation andmetastasis of NPC cells. Down regulation the level of Annexin A1promotesthe growth, proliferation and metastasis of NPC cells.
