Chapter1: The significance and headways trend of the health laboratorytechnologies for norfloxacin (NFLX) and ofloxacin (OFLO),malachite green (MG)and crystal violet (CV) were introduced respectively. The basic principles for threedetermination methods of the health laboratory technologies for them were alsointroduced briefly in this topic.Chapter2:In HAc-NaAc buffer solution, the fluorescence characteristics of NFLXand OFLX in the presence of Al3+as an enhancer were studied. A novel fluorescencemethod for the simultaneous determination of trace NFLX and OFLX in aquaticproduct was proposed based on combination of the synchronous fluorescencetechnology and the dual-wavelength method. The samples without separation candirect simultaneous determination.The measurement of the synchronous fluorescence spectra was carried out withwavelength difference of△λ=170nm for synchronous scanning. There were thelinear relationship between the synchronization fluorescence intensity difference (bythe dual-wavelength method) and the concentration of NFLX and OFLX in solutionusing Al3+as an enhancer. Under the optimized experimental conditions, selectedrespectively wavelengths446nm and523nm as the measurement wavelength and thereference wavelength for NFLX determination. There is a good linear relationshipbetween the synchronous fluorescence intensity difference of the solution with theconcentration of NFLX in concentration range of10-600μg/L, and the detectionlimit was0.29μg/L. When OFLX was mensurated, the determination wavelength andreference wavelength were467nm and431nm, respectively. The concentration range of the linear relationship between the synchronous fluorescence intensity differenceand the concentration of OFLX was20-1200μg/L. The detection limit was0.77μg/L.The method is simple and has the high accuracy, precision and the low detection limit.The method has been applied to determine the contents of NFLX and OFLX inaquatic product samples with the recoveries of95.9%-101.0%and96.3%-100.2%,respectively. The RSD were less than1.41%and2.51%, respectively. The sensitizingmechanism of Al3+was preliminarly discused.Chapter3: In this chapter, a new method was established for simultaneousdetermination of MG and CV, based on the combination of cloud point extraction(CPE) and ultra-violet absorption dual-wavelength method. This new method canenhancement the determination’s precision and selectivity compare with the commonmethod.In phthalate-HCl buffer solution, MG and CV can be extracted and enrichmentfrom the complex samples solution by Triton X-114cloud point extraction method,which result in the absorbance significantly enhance of MG and CV. selectedrespectively wavelengths624nm and512nm as the measurement wavelength and thereference wavelength for MG determination. There is a good linear relationshipbetween the absorbance difference of the solution with the concentration of MG inconcentration range of1.0×10-8-1.0×10-6mol/L, and the detection limit was2.89×10-9mol/L. When CV was detected, the determination wavelength and referencewavelength were593nm and649nm, respectively. The concentration range of thelinear relationship between the synchronous fluorescence intensity difference and theconcentration of CV was in the range of1.0×10-8-1.0×10-6mol/L. The detection limitwas1.86×10-9mol/L. The method is simple and has the high accuracy, precision andthe low detection limit. The method has been applied to determine the contents of MGand CV in aquatic product samples with the recoveries of94.4%-96.2%and93.1%-93.5%, respectively. The RSD were less than3.66%and4.04%, this method hasmany advantages which simple, high accuracy and high precision,and has some valuefor the detection of MG and CV in real sample.
