Aims To determine the role of receptor component protein (RCP) in Calcitonin gene-related peptide (CGRP) signal transduction in action of VPO1in VSMCs induced by Angiotensin Ⅱ.Methods Western blotting was used to determine the protein expressions of RCP and VPO1. RT-PCR was used to detect the mRNA expression of RCP and VPO1. RCP-specific small interference (siRNA-RCP) was used to silence oligonucleotide sequence.Results CGRP up-regulated the expression of RCP in a concentration and a time-dependent manner in VSMCs while down-regulated the expression of RCP in Ang Ⅱ induced VSMCs (P<0.05); siRNA-RCP successfully decreased RCP protein expression by80%in48h (P<0.05); in siRNA-RCP VSMCs, CGRP down-regulated the expression of RCP and inhibited the expression of RCP induced by Ang Ⅱ (P<0.05); CGRP up-regulated the expression of VPO1while inhibited the expression of VPO1induced by Ang Ⅱ in VSMCs(P<0.05), and CAT, H2O2scavenger, enhanced the inhibition effect of CGRP (P<0.05); in siRNA-RCP VSMCs, CGRP also inhibited the expression of VPO1induced by Ang Ⅱ in low levels (P<0.05)Conclusion RCP mediated the regulation of CGRP on VPO1in Ang Ⅱ-induced VSMC.
