Aims:To establish the model of rats with left carotid artery injured by Balloon, deducethe effects of TSG on the intimal proliferation in rats with vascular injury throughobserve the intimal proliferation; and study the effects of TSG on ERK1/2throughtest the expression of ERK1/2.Methods:To choose54male wistar rats from the Animal Center of the Nanhua University,randomly divided them into the sham-operated group, the injury group and the TSGgroup with18in each, and the model of left carotid balloon injury are established inthe later two groups.The left Caro-tid tissues of the three groups are harvested7、14and21days after the in jury with6rats at each time point. A part of tissues areused to HE staining,observed the intimal proliferation and the VSMC proliferation byoptical microscope.Other tissues are taken to detect the expression of pERK1/2by themethod of Western Blot and immunohistochemical staining.Results:1.The Result of HE staining:the intimal proliferated obviously in the injurygroup and the TSG group compared with the sham-operated group at each timepoint,show that the balloon injury could make the intimal proliferated;Compared tothe injury group,the intimal proliferation reduced significant in the TSG group,demonstrated that TSG can significantly lighten the intimal proliferation after theartery injured by Balloon. 2.The expression of pERK1/2detected by the method of immunohistochemicalstaining and Western Blot:The expression of pERK1/2increased obviously in theinjury group and the TSG group compared with the sham-operated group at each timepoint,explain that the balloon injury could increase the expression of pERK1/2(p<0.05);Compared to the injury group,the expression of pERK1/2reducedsignificant in the TSG group(p<0.05).Certified that TSG can downgrade theexpression of pERK1/2.Conclusion:1.TSG significantly attenuate intimal proliferation of the arteries after ballooninjury.2.The mechanism may be related to its inhibitory effect on the expression ofERK.
