Background:Atopic dermatitis (AD) is a relapsing and chronic form of inflammatoryskin disorder characterized by genetic and environmental factors. AD affecting at least15%-30%of children and2–10%of adults in industrialized countries.The pathogenesisand etiology of atopic dermatitis is not clear. The results of studies show that the geneticfactors are the main susceptibility factors to AD, and AD is a complex disease.In thepast years, researches mainly used genome-wide linkage studies, candidate genesstudies or other research methods to search AD susceptibility genes and many progresswas made. After2005, the completion of the Human Genome Project (HGP) and theInternational Haplotype Mapping Project (HapMap) provides significant biologicalinformation for research complex disease susceptibility genes. AD is geneticallyheterogeneous among different ethnic populations. Many studies were performed in theCaucasian population. But it is not clear whether these susceptibility genes/SNPs havethe same function in Chinese population.Therefore, search susceptibility genes of AD inChinese population are necessary.Objective: To screen for susceptibility genes of atopic dermatitis and provide importanttheoretical basis for the pathogenesis, diagnosis and prediction of atopic dermatitis inChinese Han population.Methods:8SNPs were genotyped by using the Sequenom MassARRAY platform in2,205cases and2,208controls of Chinese Han population. Results:We found that the SNP rs10056340(P=0.003,OR=1.188, OR=1.188,95%CI=1.058-1.335)and rs2155219(P=0.004,OR=0.884,95%CI=0.812-0.962)weresignificantly associated with the risk of AD. The study suggested that the SNPrs2155219might not only play important roles in the development of AD, but alsocontribute to the complex phenotypes of AD.Conclusions:Our results confirmed the association between rs10056340、rs2155219and atopic dermatitis in Chinese Han Population.The study indicated the SNPrs10056340not be associated with some complex phenotypes of AD: age、associateddisease、disease severity、clinical subtypes、family history.For SNP rs2155219,the studysuggested that it might not only play important roles in the development of AD, but alsocontribute to the complex phenotypes of AD.
