| Objective1. To investigate the influence of blocking the signal transduction pathway of ERK onIFN-induced antiviral proteins in HepG2cells.2. To evaluate the the relationship between those antiviral proteins expression and signalpathways in HepG2.2.15cell.Methods1. HepG2cells were treated with IFN-α and ERK inhibitor PD98059, and the mRNAlevels of MxA,2’-5’OAS, PKR, STAT1, STAT2, IRF9and SOCS3were assessed byRT-PCR after treated with IFN-α and PD98059. Meanwhile, the expression of STAT1,STAT2and SOCS3proteins were detected by Western blot.2. The HepG2.2.15cells were treated with IFN-α (IL-10or IL-12) and ERK inhibitorPD98059or STAT1inhibitor Fludarabine, and the mRNA levels of2’-5’OAS and PKRwere assessed by RT-PCR. Meanwhile, the expression of STAT1and ERK proteinswere detected by Western blot. To evaluate the the relationship between those antiviralproteins expression and signal pathways in HepG2.2.15cell.Results1. With the ERK signaling pathway of HepG2cells pre-blocked，the expression levelsof STAT1, STAT2and MxA,2’-5’OAS, PKR, STAT1, STAT2, IRF9mRNA wereenhanced significantly. However, the expression of SOCS3and its mRNA were down-regulated.2. IL-10, IL-12and IFN-a activated JAK-STAT and Raf-MEK-ERK signal pathways.IFN-a up-regulated the expression of2’,5’-OAS and PKR. IL-10and IL-12increasedmRNA expression of2’,5’-OAS, however, it had no influence on PKR. Furthermore,2’,5’-OAS and PKR expression were downregulated while JAK-STAT signal pathwaywas blocked by Fludarabine. What’s more,2’,5’-OAS was up-regulated afterRaf-MEK-ERK signal pathway being blocked by PD98059.Conclusion ERK signal pathway is involved in the antiviral activity of IFN-α throughdecreasing the expression of SOCS3in the HepG2cells.It is indicated that the antiviralproteins expression of2’,5’-OAS and PKR are up-regulated by JAK-STAT signalpathway, and the Raf-MEK-ERK signal pathway down-regulate the expression of2’,5’-OAS while has no significant effect on PKR. |
