Effects And Mechanisms Of Paeoniflorin-6’-O-benzene (CP-25) On Mice With Collagen-induced Arthritis

OBJECTIVEAccording to the changes that such as secondary paw swelling, polyarthritis index,andhistological morphological of collagen-induced arthritis （CIA） mice The Effects andmechanisms between Paeoniflorin^-6’-O-benzene sulfonate（CP-25）and Paeoniflorin（Pae） or total glueosides of paney （TGP） were compared on the model of CIA. Tostudy the effect of CP-25on peritoneal macrophage from rats with adjuvant-inducedCIA, To study the effect of CP-25on proliferation of T、B lymphocyte and thesecretion levels from mice of adjuvant-induced CIA, and study anti-inflammatoryeffects and immunomodulatory effects of CP-25on mice with Collagen-inducedarthritis.METHODSCIA was used to induce typeⅡcollagen in mice.Therapeutic treatment of CP-25(8.725,17.5,22.5,35,45,70,90,140,180mg·kg^-1),Pae(35,70,140mg·kg^-1),TGP(35,70,140mg·kg^-1),Prednisone (5mg·kg^-1Pre)（d24-d44）were given afterimmunization by interagastric administration. Hind paw volumes of mice weremeasured by volume meter.Arthritis index,spleen index were recorded.TNF-a,IL^-1,PGE2produced by thymus PMφ were measured by ELISA IL-2, IL-4,IL^-17, IL^-10,TGF-betalevel produced by thymus T and B cells was measured by ELISA.Pathological changes in synovium of joint were observed by light microscope.The proliferation of T and B lymphocytes were assayed by using MTT assay.RESULTS1. Mechanisms of effects of CP-25on CIA miceCP-25(22.5,35,45,70,90,140,180mg·kg^-1,d24-d44) suppressed significantly thepaw swelling as well as, down-regulated the index of polyarthritis, spleen index ofthymus T cells and proliferation of T and B lymphocytes and improved. The articularhistopathological changes in CIA mice. These suggested that CP-25had therapeuticeffects on CIA mice.2. Anti-inflammatory effects and Immunomodulatory of CP-25on CIA miceCompared with model, CP-25(22.5,35,45,70,90,140,180mg·kg^-1)was shown torelieve swelling of joint to some extent and the Inhibition of CP-25were moresuperior to those of TGP and Pae. Mean while, CP-25also suppressed theover—production of TNF-a, IL^-1 and PGE2. Compared with Pae and TGP group, theconcentrations of IL-4, IL^-10and TGF-β1were elevated significantly, whereas theconcentrations of IL-2and IL^-17were reduced significantly of CIA mice by CP-25.CONCLUSION:1. CP-25has good anti-inflammatory effect and immunomodulatory effect.2.CP-25has antinflammatory effect of which the mechanism may be related to themodification of the abnormal immunological function of PMφfrom CIA mice．3. The therapeutic effects of CP-25on models of CIA were more superior to those ofTGP and Pae at the some conditions.
