chitosanoligosaccharide(COS) is the only alkalescent polysaccharide in nature, it is known to posess many kinds of biological activity, such as antibacterial, anti virus, antitumor and immunoenhancing effects and so on. in addition, COS is easy to dissolve in water, wide variety of source and the preparation processs is simple and feasible, so it has a broad prospect of application in food industry. In this sduty, we research the immunoenhancing and antitumor effect of three different molecuLar weight COS(LMCOS, MMCOS, HMCOS) prepared by our own lab. The object is not only to validate the biologic activities of COS but also provide a theory to support the development of commercialization. The resulte of the experiment is as follows:The inhibition ratio to S180tumor cell of2.5mg/mL LMCOS can achieve82.31%, the inhibition ratio to S180tumor cell of MMCOS and HMCOS can achieve76.72%and71.35%respectively. The inhibition ratio to S180tumor in vivo of low,middle and high dose LMCOS can achieve31.04%,29.88%,and34.05%respectively. The inhibition ratio to S180tumor in vivo of high dose LMCOS, MMCOS and HMCOS can achieve72.52%,59.95%and81.86%respectively. LMCOS can significant (P<0.05) prevent the tumorigenic.When the concentration of LMCOS ranges from10μg/mL to1mg/mL, it can increase the phagocytosis function of mouse peritoneal macrophage(PMΦ), when the concentration is10μg/mL and100μg/mL, the differeces between experimental group and control group are considered to be statistically significant(P<0.05). COS of low (250mg/kg.bw), middle (500mg/kg.bw), high (1000mg/kg.bw) does can exceeding significant(P<0.01) increase Delayed Hypersensitivity (DTH) through intragastric administration. In addition, different dose of COS can exceeding significant (P<0.01) increase the phagocytic index, DTH, thymus index in immunosuppression mouse, high dose LMCOS can significant(P<0.05) increase the spleen index in immunosuppression mouse; Different dose of COS can increase the level of IL-12、IFN-r in serum and the level of lactate dehydrogenase(LDH) and acid phosphatase(ACP) in spleen. The above finding point out that LMCOS can enhance the immune function, can protect the immune system from Cy caused immunosuppression; Different molecuLar weight COS have strong cytotoxicity to S180tumor cell, they also can inhibit the S180tumor in vivo, when combine with Cy the inhibition is increased, this indicant that COS have a good adjunctive therapy effect besides can prevent the tumorigenic.
