Anti-tumor Activity Screening Study Of Zhu-li From Bamboo Native Yunnan

OBJECTIVE To investigate the anti-tumor Activities of MJZ-ZL in vivo and in vitro. METHODS Antiproliferative activity against human hepatic carcinoma cell HEPG2and SMMC-7721,human gastric carcinoma cell SGC7901and MKN28,human colorectal cancer cell HCT116,human colon cancer cell LOVO,human bladder carcinoma cell BIU87,human cervical carcinoma cell HELA,human breast cancer cell MCF7, human leukemia cell K562, myeloma cell U266, human melanoma cellA375, was determined by MTT assay; Taken the transplantable tumor models of H22and S180-bearing mice as models, then the inhibition ratios of tumor growth and indexes of spleen and thymus were caluclated. The serum tumor necrosis factor-a (TNF-a) and interlekin-2(IL-2) were determined with ELISA method. Preliminary discussion in vivo antitumor mechanism of MJZ-ZL. RESULT In vitro MJZ-ZL had certain inhibitory effect on SMMC-7721、 SGC7901、 MKN28、 SPCA-1、 HCT116、LOVO、BIU87、 HELA、 MCF7、 K562、 A375cells, which were comparatively obvious inhibitory effect on SMMC-7721、 SGC-7901、 MKN28、 SPCA-1、HELA cells, and50μL/mL MJZ-ZL had the inhibition rate of70%or more; the quantity of drug concentration and inhibition has a certain effect, but MJZ-ZL has no obvious inhibitory effect on A549and HEPG2; In H22-bearing mice, pretreatment and treatment of MJZ-ZL inhibited the growth of transplanted tumor with the inhibitory ratios of29.57%、18.41%, respectively and the indexes of thymus were2.90、1.99, the indexes of spleen were8.09、7.74, respectively. The concentrations of TNF-a and IL-2were61.98pg/ml and86.22 pg/ml. In s180-bearing mice, pretreatment and treatment of MJZ-ZL inhibited the growth of transplanted tumor with the inhibitory ratios of29.23%、21.61%, respectively and the indexes of thymus were3.10、2.84the indexes of spleen were8.02、9.89, respectively. The concentrations of TNF-aand IL-2were73.45pg/ml and94.27pg/ml. CONCLUSION1. MJZ-ZL inhibits the proliferation of SMMC-7721、 SGC7901、 MKN28、 SPCA-1、 HCT116、LOVO、 BIU87、 HELA、 MCF7、 K562、 A375cells;2. In vivo, pretreatment and treatment of MJZ-ZL could exhibit an inhibitory effect on the growth of transplanted H22and S180tumor,3. MJZ-ZL could promote activities of TNF-a and increase IL-2induced level4. MJZ-ZLwas suggested to increase thymus index spleen index and prlong the survival time of tumor-burdened mice. One of anti-tumor effects by MJZ-ZL may be mediated by increasing immune function.
