[Objective] To investigate the effect of distraction force at different time points on the expression of F-acti、Girdin、Akt in cultured human periodontal ligament fibroblasts(HPLF)and to discuss the mechanical signals transduction pathways in human periodontal ligament fibroblasts;to reveal the remodeling of periodontal tissue as to the mechanical effect;to further clarify the conversion mechanism and the adaptability of periodontal ligament flbroblasts during orthodontic force and molecular mechanisms of mechanical signals transtruction in cell for theoretical basis.[Methods] Took middle one part of three periodontal tissue on root,with the method of improved tissue adhering, isolation and purification of cells through the passage and observation of cell morphology and immunocytochemical identification of cell sources, got the follow-up required experimental cell. With Forcel four-point bending strength device, used0.5Hz、5,000μ strain dynamic distraction stimulation against HPLF at0hours,2hours, and6hours, used immunofluorescence confocal laser scanning microscopy, Western blot to detect the protein expression changes of F-actin, Girdin, Akt in human periodontal ligament fibroblasts.[Results]①Using the method of improved tissue adhering,we have successfully cultured and subcultured the Human periodontal ligament cells(HPLF).The cultured cells showed typical characteristics of HPLF.②Immunofluorescence observation:0hours,cytoplasm could be seen looming green fluorescent;2hour,6hour cell cytoplasmic could be obviously seen green fluorescence and fluorescence intensity increased With the extension of time.③Western blot test results:the team which was given dynamic distraction stimulation, mprotein expression of F-actin, Akt, Girdin increased in turn With the extension of time0hours,2hours and6hours.Excepted for Akt in hours of2and6increased no statistically significant differences, the other team had significant changes comparised with each other. There was a significant difference between the distraction group and the control group.[Conclusions]①We have cultured the HPLF successfully Using the method of improved tissue adhering,Through the passage of primary cell, isolation and purification, took generation of6-8of HPLF as a source of experimental cell more appropriately.②F-actin, Girdin, Akt had participated in the mechanical stress signal transduction processes in human periodontal ligament fibroblasts.③F-actin,Girdin and Akt may as the main regulation factor involved in orthodontic periodontal tissue reconstruction.
