Objective:To explore hepatic fat content and related risk facters in various glucose metabolise.Methods:1. A total of200subjects including impaired glucose regulation (IGR)(n=46),newly diagnosed type2diabetes (NT2DM)(n=64) and normal glucose tolerance (NGT)(n=90) were recruited in Tianjin during Mar.2011to Feb.2012. All subjects have no diabetes history;no severe kidney diseases and renal dsyfunction(Cr>115μmol/L).no tumors;no severe psychiatric disorders;not used drugs which maybe affect glucose metabolic such as glucocorticoids,thyrotroxine,thiazine before and under one month investigation.no chronic viral hepatitis,no autoimmune liver diseases,drug-induced liver disease and other liver diseases due to hereditary diseases.no abundance alcohol drinking(alcohol intake:male<20g/d, female<10g/d).All subjects have no hypoglycemic therapy,and have no changes of types and doses of lipid and hypotensive drugs the last one month.2. Records the following informations of the three groups:(1) Demographic data:Age, Sex;(2) General physical examination informations:①height, weight, and body mass index was calculated (BMI=weight/height2,kg/m2);②systolic blood ptessure (SBP), diastolic blood ptessure (DBP);(3) Laboratory parameters:①Biochemical indicators: alanine aminotransferase(ALT),aspartate transaminase(AST),alkaline phosphatase (ALP),gamma-glutamyl transpeptidase(y-GGT),serum uric acid (SUA), creatinine (Scr), urea nitrogen(BUN),triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C) was calculated;②fasting plasma glucose (FPG),2h plasma glucose (2hPG);(4)Acquisition liver B ultrasonic image,and calculate the liver fat content.Results:1. General data shows,age,waistline,BMI,SBP,DBP were evidence elevated in IGR and NT2DM groups(P<0.01),compared to NGT group,there was no statistice difference between IGR and NT2DM groups;TG,TC,LDL-c were elevated obviously in NT2DM group(P<0.05),compared to NGT and IGR groups,HDL-c was descend gradually with disturbance of carbohydrate metabolism,but there have no difference among three groups;FPG,2hPG were eleveted acompany with disturbance of carbohydrate metabolism(NGT<IGR<NT2DM,P<0.01);2. Prevence of NAFLD and hepatic fat content were eleveted with disturbance of carbohydrate metabolism(P<0.01);3. As whether have got dysglycemia dependent variable,other clincal datas independent variable,binary Logistic regression shows,age,hepatic fat content were independent risk factors (P were0.007,0.011,respectively);4. Correlation analysis shows,liver fat content positively correlated with SBP,DBP, BMI,waistline,WHR,FPG,2hPG,TC,TG,LDL-c,ALT,AST,ALP,γ-GGT,and correlation coefficient were0.231,0.286,0.412,0.456,0.328,0.214,0.269,0.239,0.263,0.190,0.369,0.235,0.155,0.186(P both<0.01); and negatively correlated with HDL-c,and correlation coefficient was-147(P<0.05);5. Liver fat content as the dependent variable and gender,age,SBP,DBP,BMI,waitline,WHR,FPG,2hPG, TC,TG,LDL-c, ALT,AST,ALP,γ-GGT as the independent variable for the linear multivariate regression analysis, the results show the waistline,ALT for regression equation of liver fat content,(β were0.368,0.193, respectively)(P<0.05,0.01, respectively).Conclusion:Prevence of NAFLD and hepatic fat content was eleveted with disturbance of carbohydrate metabolism,and hepatic fat content was independent risk factor of glucose dysregulation.Hepatic fat content was associated with SBP,DBP,BMI,waistline,WHR,FPG,2hPG,TC,TG,LDL-c,ALT,AST,ALP,y-GGT,HDL-c,and waistline, ALT play important role in process of NAFLD.
