Objective:We performed an analysis of miRNAs expression profile in peripheral blood of psoriatic patients and healthy controls by miRNA chip technology. We also predicted the target genes of the differentially expressed miRNAs and analysed their molecular fuctions in order to determine whether miRNAs are involved in the pathogenesis of psoriasis.Methods:According to the selected standards, we collected3patients of psoriasis vulgaris in active phase and2healthy controls. Then we extracted total RNA and carried on quality and purity assessment. Comply with the requirements of Affymetrix miRNA chip technology, we performed fluorescent labeling and hybridization in accordance with the standard test process. Then the hybridized chips were scanned by Affymetrix GeneChip Scanner3000, and the data were read and processed by Affymetrix GeneChip Operating Software Versionl.4. Differentially expressed RNA were selected according to the prescribed standards(the q-value(%)≤5and the fold chang≥2). Target genes of these differentially expressed miRNAs were predicted by Human Target Scan5.1database and their molecular fuctions were analysed by Pathway and Go classification database of MAS system.Results:We identified5miRNAs that were consistently differentially expressed between experimental group and healthy controls, which, hsa-miR-363hsa-miR-30e、hsa-miR-192up-regulated, and hsa-miR-423-5p、hsa-miR-720down-regulated. And5differentially expressed miRNAs sequence length are all concentrated in17to23nucleotides. The total number of the target genes is493. The Pathway classification results show that the biological processes of these target genes are involved in multiple signaling pathways, the development of a variety tumors, apoptosis, the interaction of cytokine, et al. The GO classification results demonstrate that the molecular functions of these target genes are enzyme activity, receptor activity, cytokine activity, et al. And the biological processes are involved in DNA replication, RNA degradation, apoptosis, angiogenesis, inflammatory response, immune response, the proliferation and differentiation of immune cells, et al.Conclusion:There are differentially expressed miRNAs in the peripheral blood of patients with psoriasis, which, hsa-miR-363、hsa-miR-30e、hsa-miR-192up-regulated, and hsa-miR-423-5p、hsa-miR-720down-regulated. The total number of the target genes is493. Our experimental results suggest that these miRNAs may contribute to psoriasis pathogenesis by modulating their target genes. Thus, our research may reveal a new molecular level in the pathogenesis of psoriasis.
