Objective:In this study, we investigated the antifibrotic potential of Oligomeric Proantho Cyandins (OPC) on LPS-stimulated HSC-T6cells in vitro experiment.Methods:Cell viability was detected with the3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Hepatic stellate cells (HSCs) were treated with OPC (5,10, and20μg/ml) with or without LPS (1μg/ml) for24h. Western blotting were performed to detect type I collagen (Collagen-Ⅰ), a-smooth muscle actin (a-SMA), tissue inhibitors of metalloproteinases-Ⅰ (TIMP-1), metalloprotease-13(MMP-13), Phosphatidylinositol3-kinase (PI3K) and AKT protein levels. Nuclear factor-kappa B (NF-κB) translocation was detected by immunocytochemistry staining.Results:Our findings demonstrate that OPC could inhibit cells viability of HSC-T6cell, decreased expression of collagen-Ⅰ, α-SMA, TIMP-1on LPS-induced HSC-T6cells. OPC also significantly inhibited LPS-stimulated PI3K, AKT, attenuated LPS-teiggered NF-κB translocation.Conclusion:Our findings implied demonstrate that OPC inhibited LPS-induced expression of collagen-I, a-SMA, TIMP-Ⅰ via suppressing of the PI3K-AKT and NF-κB signaling pathway.
