Multiple Polymerase Chain Reaction In The Diagnosis Of Infectious Keratitis And Its Significance

Purpose:Select the various causes of keratitis patients, experiment with Corneal epithelial or conjunctival secretions for DNA extraction and PCR amplification. Analysis of its results.Research multiplex polymerase chain cause keratitis in clinical diagnosis diagnostic significance and value.Methods:Select keratitis patients with a total59cases.Considered keratitis29cases of bacterial infection (group I), considered16cases of fungal keratitis infections (group II), considered herpes simplex virus keratitis in14cases (group III),20cases of normal eyes (group â…£). Added bacterial universal primers, herpes simplex virus and fungal universal primers to all subjects corneal epithelial extracted DNA,Amplification for MPCR.Observe the results with Gel electrophoresis gel image analysis system.Results:(1)In group â… ,510bp has20, while510bp,310bp has1eye,positive rate of72.4%, microbial culture results positive rate was72.4%.Using X2test detected no significant difference.(X2=0.000, P=1.000).(2)In group II,310bp have8eyes, while amplified52Obp and31Obp has none,the positive rate was50.0%, fungal microbial culture results positive rate was52.9%,Using X2test detected no significant difference.(X2=0.29, P=0.866).(3) The comparison of infectious keratitis three group with group IV (control group) of MPCR positive rate, the results were statistically significant differences (P<0.001).(4)Clinical diagnosis of infected fungi, bacteria, herpes simplex virus patients, with MPCR detected corresponding number of cases for the pathogen was in34cases, consistent rate of57.6%, two sets of data using X2test detected a significant difference (X2=31.72, P<0.001).Conclusions:1. MPCR method is an effective and reliable method at infectious keratitis diagnosis and differential diagnosis.2. Compare With the traditional culture of bacteria and fungi,MPCR method has Advantage of fast and economical,its has a good application prospect.
