[Objective] To research the preventive effect of astragaloside on ischemia-reperfusion injury of skeletal muscle and discuss the possibility biochemistry mechanism.[Method] Wistar rats (70,weight180-200g) were used to determine whether astragaloside existed toxicity effect.And Wistar rats (male, weight180-200g) were used to determine activity of creatine phosphokinase (CPK) and superoxide dismutase (SOD).20rats were divided into experiment group with astragaloside and control group with distilled water. Keeping on one week one time every day observe the normal condition of rats to detect the toxicity effect. Wistar rats were divided into three groups. Modle group:animals were given astragaloside by gavage for one week before ligaturing the right arteria cruralis for4hours, flood again, intragastric administration with10%glucose1ml/100g).Sham operation:arteria cruralis was only separated and intragastric administration with10%glucose. Astragaloside groups:It consisted of three groups low dose group (30mg/kg), middle dose group(60mg/kg), large dose group90mg/kg).Others were administration as well as the control groups.Finally, determine the activity of CPK and SOD in blood serum to research the effect of astragaloside on ischemia-reperfusion injury of skeletal muscle.[Results]1. Effects of astragaloside on the rat normal conditions.After intragastric administration with astragaloside for seven days, the behavour,feed, fur, breath, motion,weight of rats were all normal and exist non-toxic else. Watching the change of visceral organs after seven days, did not discover abnormal. Astragaloside group was not significant different compaired with control group (P>0.05).2. Effects of astragaloside on CPK in blood serum of rats.CPK in the serum in model group were obviously increased compared with sham operation group (P<0.05); Astragaloside with big dose and middle dose can significant degrade compaired with model group,(P<0.05); Though CPK in low dose groups reduced, it was not significant.3. Effects of astragaloside on SOD in blood serum of rats.SOD in the serum in model group were obviously decreased compared with sham operation group (P<0.05); But astragaloside with big dose and middle dose can significant increased compaired with model group,(P<0.05); Though SOD in low dose group also increased, it was not significant.[Conclusion]1. The astragaloside is obvious non-toxic effect.2. The astragaloside can prevent ischemia-reperfusion injury,it is possible to do with reduced CPK and increased SOD.
