Objectives:To investigate the expression and clinicopathological significance of the Ezrin protein in colon cancer.Materials:Total72cases of paraffin-embedded colon lesion blocks, including10cases of normal colon tissues and62cases of colon cancers, were selected from Department of Pathology, Yanbian University Hospital and Yanbian Tumor Hospital. In the colon cancer,21of well differentiated carcinoma,26of moderately differentiated carcinoma,15of poorly differentiated carcinoma;34of lymph node metastases,28of without lymph node metastases; In clinical pathological stages,7of stage Ⅰ,27of stage Ⅱ,24of stage Ⅲ,4of stage Ⅳ.Methods:Ezrin gene protein expression level was detected in colon cancers and normal colon tissues by using EnVision method of immunohistochemistry, and the clinicopathological significance of Ezrin protein in colon cancer was also analyzed.Results:Immunohistochemical staining showed that the expression of Ezrin protein was low level in normal colon tissues, with only20.0%(2/10) positive rate, the positive and the strongly positive rates of Ezrin protein were83.3%(56/62) and66.7%(42/62) in colon cancers (P<0.01). The positive rate of Ezrin protein was71.4%(15/21) in well differentiated carcinoma,53.8%(14/26) in moderately differentiated carcinoma,86.7%(13/15) in poorly differentiated carcinoma, no significant difference. Immunohistochemical staining also showed that the positive rate of Ezrin protein was differently among the clinical pathological stages in colon cancers,91.7%(22/24) of III stage and100%(4/4) of IV stage were significantly higher than42.9%(3/7) of I stage and48.1%(13/27) of II stage (P<0.01). The less5years free survival rate of Ezrin protein was92.3%(24/26), it was higher than50.0%(18/36) of5years and over5years (P<0.05) Conclusions:The detection of Ezrin protein might be a useful marker for early diagnosis and prognosis of colon cancer. And Ezrin gene might be the new target for colon cancer therapy.
