Objective:The objective of this study is to clarify protective effects of Ginkgo biloba da mo injection on human vascular endothelial cells induced by hypoxia-reoxygenation injury by observation of morphology, MTT assay, assaying the cotent of NO and MDA and activity of SOD in the fluid supernatant fluid of cultured cells,to probe the inherent mechanism of the drug on the physical therapy of coronary artery ischemia reperfusion injury,further providing comprehensive theoretical basis for the protection on blood vessel endothelium cell which is undergoing coronary stent implantation(the cell of the block vessel which is recanalized), guiding clinic medicine and providing scientific envidence for designing therapeutic schedule and developing the effective clinic medicine on the resistance to reperfusion injury.Methed:This experiment adopted to HUVECs which were cultured from generation to generation in vitro. Vascular endothelial cells were divded into three groups:â‘ normal group:normal VECsâ‘¡hypoxia-reoxygenation group:VECs which were treated with1mM sodium dithionite and continued to be cultured for24hours by adding whole DMEM.â‘¢Ginkgo biloba da mo group:VECs were continuously cultured with different concentrations of Ginkgo biloba da mo after hypoxia. The morphological changes of VECs in each group were observed under the inverted microscope. The optical density (OD) value of cells was detected by MTT colorimetric method, the content of MDA and NO in the supernatant fluid of cultured cells was detected by ELISA, and the activity of SOD was detected by ELISA.Results:The OD value was obviously decreased in the hypoxia-reoxygenation group, but the one was increased in the normal and Ginkgo biloba da mo injection group.The OD value had significant difference (P<0.05) among normal group, Ginkgo biloba da mo injection group and hypoxia-reoxygenation group by MTT assay. The content of MDA was increased and the content of NO was decreased in the hypoxia-reoxygenation group, in contrast, the content of MDA was decreased and the content of NO was increased in the normal and Ginkgo biloba da mo injection group. The content of MDA and NO in the supernatant fluid of cultured cells also had significant difference among the hypoxia-reoxygenation group, normal group and Ginkgo biloba da mo injection group (P<0.01).The activity of SOD was declined in the hypoxia-reoxygenation group, in contrast, the activity of SOD was rised in the normal and Ginkgo biloba da mo injection group. The activity of SOD had significant difference (P<0.01) between normal group, Ginkgo biloba da mo injection group and the hypoxia-reoxygenation group. But the normal group and Ginkgo biloba da mo injection group had no difference (P>0.05).Conclusions:The results indicate that the Ginkgo biloba da mo injection has protective effects for the HUVECs induced by hypoxia-reaxygenation injury on the resistance to oxidative stress.
