Objective CD4+T cell and its related cytokines had an important role in antuimmune diabetes. PTPN22gene (+1858C/T) polymorphism was associated with this kind of disease.This study will analyze and compare the amount of regulatory T cell and CD4+effector T cell, expression of IL-2, IL-10, IL17and IFN-gamma, PTPN22gene (+1858C/T) polymorphism in type1diabetes (T1DM), latent autoimmune diabetes in adults (LADA), type2diabetes (T2DM) patients and healthy people as control group.Methods Participants of this study include19T1DM,14LADA,20T2DM patients (age≤45years, duration of the disease≤5years, BMI≤30kg/m2) and20healthy people. The amount of regulatory T cell and CD4+effector T cell was analyzed by flow cytometry. Expression of cytokines such as IL-2, IL-10, IL17and IFN-gamma were measured by real time-qPCR. PTPN22gene (+1858C/T) polymorphism was tested with gene sequencing.ResultsPercentage of Treg and CD4+Teff cell in CD3+CD4+T cellCD3+CD4+CD25+CD127-T cell represented the Treg cell. The percentages of Treg in T1DM, LADA, T2DM and control group were6.39±1.82%,7.32±2.42%,6.62±1.69%,6.48±2.32%respectively, with no significant difference(F=0.64, P=0.592). CD4+Teff counted for93.61±1.82%,92.67±2.42%,93.37±1.69%,93.52±2.32%of CD3+CD4+T cell inTlDM, LADA, T2DM and control group respectively, with no significant difference(F=0.641, P=0.591).Relative expression of IL-2, IL-10, IL17and IFN-gammaRelative expression of IL-10in LADA group(1.35±0.54) was significantly higher than T2DM(0.98±0.41) and control group(1.0±0.34)(p<0.05). And no difference was found between LADA and T1DM group(1.15±0.3). No significantly difference of relative expression of IL-2was found in T1DM group(1.2±0.46),LADA(0.98±0.4),T2DM(1.0+0.24) and control group(1.0±0.32).Relative expression of IL-17were1.07±0.48,1.03±0.4,0.96±0.34and1.0±0.34in T1DM, LADA, T2DM and control group respectively with no statistical difference. Relative expression of IFN-gamma were1.02±0.28,1.13±0.51,1.04±0.35and1.0±0.34in T1DM, LADA, T2DM and control group respectively with no statistical difference.There was inverse correlation between Treg pecentage and IFN-gamma expression in T1DM group(rs=-0.678, P=0.001).There was direct correlation between Treg pecentage and IL-10expression in T1DM group(rs=0.773, p<0.001).PTPN22gene (+1858C/T) polymorphism analysisNo difference of PTPN22gene (+1858C/T) genotype was observed in both4groups. No correlation was founf between PTPN22gene (+1858C/T) polymorphism and expression level of IL-2, IL-10,IL-17and IFN-gamma.ConclusionRelative expression of IL-10in LADA group was significantly higher than T2DM and control group. And there was a trend that IL-10expression was higher in LADA group than T1DM which suggested that the autoimmune damage of islet beta cells was milder in LADA. Correlation between Treg percentage, IFN-gamma and IL-10expression in T1DM suggested that the balance between protective factors and destructive factors was broken which represented as the disequilibriumof IL-10and IFN-gamma. No statistical difference of PTPN22gene (+1858C/T) genotype and its correlation with IL-2,IL-10,IL-17and IFN-gamma was observed in T1DM,LADA,T2DM and control group.
