Objective: To observing the expression changes of signal transduction pathway innetwork of enteric nerve-interstitial cells of Cajal (ICC) in the model of rats with YangMing Fu Shi Syndrome, and the therapeutic effects of Da Cheng Qi Decoction (DCQD).Methods: One handred Wistar rats were randomly divided into controlgroup(n=20), Yang Ming Fu Shi Syndrome model group (n=40), and Da Cheng QiDecoction group (DCQD group)(n=40). The rats of control group were injected l.0mlof normal saline into the peritoneal cavity. The rats of Yang Ming Fu Shi Syndromemodel group were perfused Chinese herb medicine of the hot and itself dung, then1.0ml suspension of Escherichia coli was injected into the peritoneal cavity. The rats ofDCQD group were administrated by gavage with DCQD (twice a day, each one1.0ml/100g) after the Yang Ming Fu Shi Syndrome was established. After five days, theproximal10cm segment of jejunum was taken and the whole mount preparation fromeach group were studied used c-kit, protein kinase A (PKA), protein kinase C(PKC),and caveolin-1immunofluorescence double-staining with confocal laser scanningmicroscopy, and the immunohistochemical analysis was observed.Results: There was extreme expansion of gastrointestinal tract and obvious signsof intestinal obstruction in Yang Ming Fu Shi Syndrome rat induced by bacterialperitonitis; DCQD can significantly alleviate the signs of paralytic intestinalobstruction.Immunofluorescence study: Compared with the control group, the quantity andthe integrity optical density (IOD) of ICC-DMP of intestine in Yang Ming Fu ShiSyndrome model group were significantly decreased (P<0.01). The ICC-DMP networkwas significantly disrupted, and the integrity optical density (IOD) of PKA、PKC、 caveolin-1of intestine in Yang Ming Fu Shi Syndrome model group were significantlydecreased (P<0.01). Compared with the Yang Ming Fu Shi Syndrome model group, thequantity and the IOD of ICC-DMP of intestine in DCQD group were significantlyincreased (P<0.01). The ICC-DMP network was significantly recovered, and the IOD ofPKA、PKC、caveolin-1of intestine in DCQD group were significantly increased(P<0.01).Immunohistochemistry study: Compared with the control group, the quantity ofICC-DMP of intestine in Yang Ming Fu Shi Syndrome model group were significantlydecreased (P<0.01), the ICC-DMP continuity was significantly disrupted,and theintegrity optical density (IOD) of PKA、PKC、caveolin-1of intestine in Yang Ming FuShi Syndrome model group were significantly decreased (P<0.01). Compared with theYang Ming Fu Shi Syndrome model group, the quantity of ICC-DMP of intestine inDCQD group were significantly increased(P<0.01),the ICC-DMP continuity wassignificantly recovered, and the IOD of PKA、PKC、caveolin-1of intestine in DCQDgroup were significantly increased(P<0.01).Conclusion: The gastrointestinal tracts of the rats with Yang Ming Fu ShiSyndrome were significantly distented, and the syndrome of intestinal obstruction wasremarkable. DCQD can significantly alleviate the signs of paralytic intestinalobstruction.The quantity of ICC-DMP of intestine in rat with Yang Ming Fu Shi Syndromewas decreased and the network was disrupted. Signal transduction pathway of entericnerve-interstitial cells of Cajal was damaged. DCQD can protect the ICC-DMPnetwork and the ENS-ICC-DMP signal transduction pathway from destructing in ratwith Yang Ming Fu Shi Syndrome.
