ObjectiveThis study was designed to investigate the therapeutic effect of Rg1and the potentialmechanisms on cecal ligation and puncture (CLP) induced sepsis in mice.Methods1、Established cecal ligation and puncture induced sepsis mice model, survival wererecorded for7days.2、The effects of Rg1on bacterial burdens in the peritoneal lavage fluids and bloodfrom septic mice were calculated.3、The protective effects of Rg1on the tissues of liver and lung from septic micewere tested through HE stainings.4、 The influence of Rg1on the septic mice plasma cytokines production weredetected by ELISA.5、The percentage of apoptotic CD3+T lymphocytes derived from the thymus inseptic mice were calculated by AV-PI stainings.6、The effects of Rg1on the number of neutrophils from septic mice peritoneallavage fluids, on the number of CD4+T lymphocytes, CD8+T lymphocytes, CD11c+dendritic cells and CD19+B lymphocytes derived from the septic mice spleen werecalculated by flow cytometry.7、The effects of Rg1on the apoptosis of splenocytes in septic mice were detected byTUNEL staining.Results1、Septic mice treated with Rg1showed a significantly higher survival rate comparedwith CLP group(p<0.01).2、Treatment with Rg1decreased bacterial counts in both blood and peritoneallavage fluid compared with control septic mice(p<0.01).3、Rg1showed excellent protective effects against sepsis induced tissue damages.The pathological score of liver and lung in Rg1group were lower than CLP group(p<0.05).4、After adminstrated with Rg1, the levels of TNF-α, IL-10were lower than CLPgroups(p<0.01), as well as the level of IL-6(p<0.05). 5、The apoptosis percent of CD3+T lymphocytes in thymus of CLP-operated micewith Rg1were much lower than CLP group (p<0.05).6、Treatment with Rg1increased neutrophil counts in peritoneal cavity in septic micecompared with CLP group(P<0.01). as well as the number of CD4+T lymphocytes, CD8+T lymphocytes and CD11c+dendritic cells in spleen of CLP-operated mice withRg1(p<0.05).7、After Rg1adminstrated, the mean density of TUNEL+splenocytes were lower thanCLP group(p<0.05).ConclusionGinsenoside Rg1reduced the bacterial burdens, alleviated the injuries of liver andlung, and improved the survival rate eventually through suppressing the levels of plasmacytokines, increasing the counts of neutrophils in the peritoneal lavage fluids andameliorating the number of immune cells and the percent of apoptotis,...
