Purpose:1. On the basis of consulting a large number of literature at domestic and overseas, constructingPrague-dawley female rat Experimental periodontitis model,observing the process ofpathological changes in periodontitis and the change in the form of Periodontal tissue indifferent level Estrogen,Provide the experimental conditions for studying the influence ofEstrogen in process of periodontitis.2. Observing the changes of periodontitis female rat under different level Estrogen and theprocessing of HSP27in periodontitis.Investigation HSP27in any role of periodontitisprocessing.Research methods:1. The way to Consult the rat-model of endogenous Hormone deficiencies:Specific PathogenFree Sprague-Dawley female rats33,26weeks-olds, about240g each one,full dentition,normal occlusion,All health,have no disease。Randomly divided to3teams:SHAM group(11),OVX group(11),OVX+E group(11),and operation(include sham-operation orBilateral ovarian resection operation) after the Adaptive feeding (7days).the second day,theOVX+E group hypodermic injection17β-estradiol,the specification is20μg/kg,once threedays。The OVX group constructs the rat-model of endogenous Hormone with a low levelwithout estrogen.2. Construct the Sprague-Dawley female rats of periodontitis model: the3groups aboveperiodontal ligation after the surgery ended in3weeks. Choose a left maxillary first molars,ligate with Orthodontic ligature, and settled subgingival.The other side is contrast, without anyoperation. Observe the clinical manifestation about periodontitis at the2,4,6weeks after theended of operation, and kill3of each group to acquire Bilateral maxillary alveolar bone beforegosub their blood.3. Detection stage: chemical examination the blood of angulus oculi medialis to observe the ofestrogenic level; observe histopathological changes of paradentium after decalcification,embedding, tissue slice, H.E dyeing process of Bilateral maxillary alveolar bone, and observethe expression of HSP27under electronic scanner microscope after Immunohistochemicalstaining according add HSP27antibody. Result:1. Weight change:there were no significant differences between the groups, in the first week andafter surgery. Increment of the groups which had the sham-operation or bilateral ovarianresection operation is higher than the others groups, the weight of OVX group is growing faster.The rise of weight was no significant difference between SHAM group and OVX+E group afterthe sham-operation or bilateral ovarian resection operation from the second week until the end.2. The result of structure Endogenous estrogen deficiency rat model: After the routine testinner canthus blood of all groups which periodontal operation ended2,4,6weeks,the resultsshow that,there was no significant differences of estrogen level between SHAM group andOVX+E group at any time; the reduction of estrogen level between OVX group and SHAMgroup displayed a marked difference at any time point.The predominant lesion at autopsyshows,the uterus of OVX group are more significant atrophy than SHAM group,and the oariumcannot be found in OVX group.3. The result of construct experimental periodontitis of model rats:after all groups had shamoperation or bilateral ovarian resection operation at the3week, the subject have periodontitisligation. And then feed the rats as before. Observe the clinical manifestation about periodontitisat the2,4,6weeks after the ended of operation and HE staining section, the result shows, thegingival of all groups turned hyperemia and probe bleeding at the2week after the periodontitisligation ended, and peritonitis aggravated of all group at the2,4,6weeks, OVX group is worsethan the others group. All shows, the experimental peritonitis model is done.4. The result of HSP27expressed:In the process of the development of periodontitis,the levelof estrogen influences the express of HSP27more or less.The express of HSP27in periodontalwound at affected side is higher than the normal one in the early metaphase of periodontitisdevelopment; there is no significant differences between affected side and normal one at theexpress of HSP27at the terminal periodontitis.The fact of the influences shows,the lower thelevel of estrogen level in the early metaphase of periodontitis development,the higher of HSP27express of affected side than the nomal side,but at the terminal periodontitis, there is nosignificant differences between affected side and normal one at the express of HSP27.Theseresults implied that the effect of the estrogen influences is according to meddle in thedevelopment of periodontitis,but the mechanism remains unclear. Conclusion:1. Estrogen level may affect the weight of rats, and as time goes on, endogenous deficiency ofestrogen would increase the weight of rats.2. Estrogen level may influence peritonitis PD, the decrease of the estrogen level mayaggravated peridentitis and increase munity, especially aggravate the absorb of alveolar bone3. Expression of HSP27is different at the PD of peridentitis. The degree of inflammationthe positive correlated with HSP27express at the early metaphase of periodontitisdevelopment,and it was negatively related to estrogen level.At the terminal periodontitis,theexpress of HSP became normal.The mechanism in the change of the express of HSP is need tobe further gone into.
