Objective To investigate the effects of Pretreatment with Etomidate on the contents of ALT、AST in plasma and MDA、SOD in hepatic tissue during hepatic ischemia reperfusion injury of dogs.And to observe the hepatic injury through light microscope.Methods20adult healthy hybrid dogs were randomly divided into4groups(n=5):Sham group(S)、Ischemia reperfusion group(I/R)、Etomidate small dosage group(E1,0.6mg/kg)、Etomidate large dosage group(E2,1.5mg/kg),dogs in all groups were injected Etomidate or NS except Sham group through the veins.The sham group only separaed porta hepatis, threading and not ligated. The model of HIRI were established by ligating porta hepatis.The biochemical indicators of ALT、 AST in plasma were measured at four time points:before blocked,30min after ischemis,1h and2h after reperfusion. And measured the content of malondialdehyde(MDA) and activity of superoxide dismutase(SOD) in hepatic tissue.Hepatic tissue was observeed by light microscope after operation.Results Compared with group S,group I/R、group E1and group E2the expression of ALT、AST in plasma were increased(p<0.05);Compared with group I/R,group E1and group E2the expression of ALT、AST in plasma were decreased significantly (p<0.01),and group E2more obviously[Reperfusion2h:ALT:2911±89U/L(I/R)、863±95U/L(E2);AST:2722±103U/L(I/R)、056±115U/L(E2)].Compared with group S.group I/R、group E1and group E2the expression of MDA were increased and group I/R the expression of SOD were decreased,group E1and group E2the expression of SOD were increased in hepatic tissuc(p<0.05);Compared with group I/R,group E1and group E2the expression of MDA were decreased significantly and the expression of SOD were increased significantly in hepatic tissue(p<0.05),and group E2more obviously[MDA:21.70±2.01nmol/mg·prot(I/R)、12.69±1.02nmol/mg·prot(E2)、SOD:191±12nmol/mg·prot(I/R)、634±26nmol/mg·prot(E2)].Light microscope showed evidently that the pathological changes of hepatic tissue were more slight in group E1and group E2than that in group I/R,and group E2was slighter than group E1.Conclusion Pretreatment with Etomidate has good protective effects on HIRI,and it is related to the control on lipid peroxidation of Etomidate.And the protective function of Etomidate large dosage group(1.5mg/kg) was more reliable.
