Objective On the basis of constructing rats in chronic cancerous pain with MADB-106rat metastatic breast cancer cells-subarachnoid puncture cathete model with, and intrathecal administration of HEK293cells, which have build, genetically modified by human preproenkephalin gene, to the rats. By means of the behavior of rats and leucine enkephalin in the cerebrospinal fluid (L-EK) content changes, in order to observe and study the analgesic effect of the genetically modified cells.Methods The healthy female SD rats were punched in rats left posterior tibial in order to inject of MADB-106rat breast cancer cells with5μl. By observing the changes made the mould parts in the X line, The40rats, which were built model successfully screened by X ray, were selected.The40female SD rats were randomly divided into transplantation group(Intrathecal injection of pLV-Ubc-hPPE/HEK293cell suspension) and the control group(Intrathecal injection of blank control), since7days intrathecal transplantation, through free walk score, thermal pain threshold and the number of spontaneous condensation foot reflex behavior observation, respectively evaluate rats induced by walking, thermal hyperalgesia and aching, to compare cell lines before and after transplantation, sensitive degree of pain in rats. Experimental ice bath after the rat cerebrospinal fluid collection, through the expression of the ria method to detect people forebrain brown peptide.Results1Transplantation group rats since25days free walk score gradually reduce, two groups of rats since21days (7days hPPE/HEK293cells) from a point in time free walk score comparison was statistically significant (P<0.001):Occurred since21days after the transplant group hot pain sensitivity decreases.25days hot pain ease gradually obvious, and the control group at this point in time, measure the heat pain threshold comparison, the difference was statistically significant (P0.001); Occurred since21days after the transplant group spontaneous contractions foot reflection times is gradually reduce, the25th day of spontaneous condensation foot reflection times compared with the control group the difference was statistically significant (P<0.001).2Detecting the rat cerebrospinal fluid in the2groups in the forebrain brown peptide original expression with the method of ria:The result shows the control group had detected a tiny amount of leucine enkephalin (L-EK), detect the content of L-EK transplantation group was obviously higher than that of control group, compared the difference was statistically significant (P<0.001), the expression amount available pg concentration said.Conclusions SD female rats with intrathecal transplantation by lentivirus transfection hPPE modified HEK293cells in rat forebrain brown peptide secretion of sheath can be sustained within the people, and chronic cancer pain caused by cancer has strong analgesic effect.
