Purpose:To discuss the effect of both infection of Antisnail plasmid and treatment of the DNA methytransferase inhibitor5-Aza-2’-deoxycitydine(5-Aza-2’-CdR) on the growth of poorly differentiated gastric cancer cell line BGC823. And the influence on the mRNA expression of Snail、E-cadherin. Method:The effect was evaluated with MTT、flow cytometry、and quantitative real-time PCR. The result was show as X±S, analied by SPSS17.0P<0.05was statistical significance. Results:Compared with the control group2.5uM、5.0uM5-Aza treated group dramatically up-regulated E-cadherin mRNA expression(P<0.05). There’s difference between0.1A and2.5A as well as5.0A, but no difference between2.5A and5.0A. Pairwise comparison between time at12h、24h、48h, there were difference separately(.P<0.05),5-Aza treated group also up-regulate Snail gene mRNA. There were difference between A0.1and A2.5, A5.0(P<0.05), at48h up-regulated Snail Gene expression significantly. Compare between before and after transfection of AntiSnail Plasmid Snail gene mRNA was decreased dramatically but E-cadherin mRNA expression was increased (P<0.05). Growth of the BGC823cell line was inhibited by the two intervention methods which also increased apoptosis rate of BGC823as dose-time dependent. Conclusion:5-Aza up-regulate the mRNA expression of E-cadherin gene and Snail gene, both are dose-time dependent. Indicate that:Methylation can double inhibit Snail and E-cad gene expression. Transfection of AntiSnail can up-regulate E-cad gene expression but down-regulate Snail gene expression,indicate that:Snail is positively correlated with methylation in early gastric cancer,The infection of Anti-Snail and treatment of5-Aza both inhibited the proliferation of poor proliferation gastric cancer cell line. Combination of them have synergistic effect.
