Backgroud:Dao-di Herbs are considered the most important part of traditional Chinese medicine and the treasure of China, which plays a decisive role in the development of medicine career. In the process of long-term medical practice,"Dao-di Herbs" has become synonymous with high quality medicinal herbs. We can put forward innovative idea that taking the medicine quality evaluation method and authentic standard as a break-through, starting from the effect, evaluating quality by effect. As we know, conventional efficacy pharmacology test is a important method to identify differences in Chinese herbs from authentic and non authentic regions. Cortex Moutan, which is originally recorded in Sheng Nong’s herbal classic, s a commonly used traditional Chinese medicine. Anhui is the major place for the producing of Cortex Moutan. Cortex Moutan which is planted in phoenix mountain of Anhui Tongling is regarded as authentic herbs for its high quality, and it is famous both at home and abroad. But Feng Cortex Moutan has no advantage in market competition, while the Bozhou cortex moutan become the biggest production in the market of cortex moutan.According to modern medicine disease surveillance, diabetes mellitus as a non infectious epidemic is spreading all over the world. In our country, the incidence of diabetes is on the rise, and patients with early onset of severe cases. Now the main methods to treat diabetes are diet, medication and exercise therapy. However, the current clinical treatments of DM are common focus on drug treatment and auxiliary in diet, while lack of awareness of exercise therapy for diabetes, and experimental study and clinical application is also not enough.Objective:1. To study the antihyperglycemic effects of Cortex Moutan from authentic and non authentic regions by copy the type2diabetic mice model. We want to provide the scientific experimental basis for clinical application, and evaluation the quality difference of herbs from authentic and non authentic regions.2. By Combining the antihyperglycemic bioactivity of Cortex Moutan water extract and exercise, then reviewe their mechanism of diabetes treatment, and provide the reference for the exercise therapy of diabetes.Methods:1. Two kinds of Cortex Moutan was respectively boiled with10times distilled water for3hours and repeated for3times. Then the extracts were filtered and the filtrates were then concentrated to dryness under reduced pressure and further freeze-dried to give the water extracts. The type2diabetic mice model was established by intraperitoneal injection of STZ and then the experimental mice were randomly divided into the model group, the group treated with Metformin hydrochloride, the group treated with authentic Cortex Moutan and the group treated with non-authentic Cortex Moutan. The body weight, the levels of fasting blood glucose, glucose tolerance and other blood biochemical indexes were determined after4weeks of the treatment. At the same time, the changes of related genes including Insr, G6pc and Gck were determined by the way of molecular biology.2. The SPF male Kunming mice adaptive feeding three days, then randomly selected8as normal control group. Other mice were fed with a homemade high fat diet for4weeks, then one-time to the mice by intraperitioneal injection of STZ solution to found the type2diabetic model. Then the experimental mice were randomly divided into4groups:the model group, the group treated with Feng Cortex Moutan, the exercise group and the combined group (treated with Cortex Moutan and exercise). The mice in Cortex Moutan group and the combined group were given Cortex Moutan water extract every day. While the mice in exercise group and combined group were running every day.4weeks later, all mice were anatomied and determined the content of GLU in serum. Lastly, the data were statistically analyzed.Results:1. The two kinds of Cortex Moutan extract both have good effects on body weight, water intake, food intake and mental state in diabetic mice. The authentic Cortex Moutan and the non-authentic Cortex Moutan water extract both could significantly reduce GLU(P<0.01), TC(P<0.05) and MDA(P<0.05), and significantly increase SOD(P<0.01)in type2diabetic mice. After dosing, two kinds of Cortex Moutan both can significantly reduce the relative expression of G6pc and can significantly increase the relative expression of GCK and Insr. Among them, the hypoglycemic and lipid-lowering effects of Feng Cortex Moutan were better than Bozhou Dan Cortex Moutan (P<0.05), whlie the antioxidant effect was worse, but the difference do not have significant (P>0.05).2. After4weeks, Cortex Moutan and exercise both make a gradual improvement in the spirit of diabetic mice, increased activity and weight gain. Compared with model group, the biochemical index of Cortex Moutan group, exercise group and combined group both has significant difference (P<0.01, P<0.05). And the experimental data of combined group compared with Cortex Moutan group and exercise group have significant difference (P<0.05).Conclusions:1. Both the two kinds of Cortex Moutan can significantly reduce the blood glucose in type2diabetic mice and can adjust the lipid metabolism and antioxidant capacity, and the data were without significant difference. The effects of authentic Cortex Moutan were better. The differences may be associated with their different production areas. Therefore, we should be combined with the different effects on selection of Cortex Moutan.2. Cortex Moutan water extract and exercise are both have good hypoglycemic effect. And combined treatment method of Cortex Moutan plus exercise has the most significantly influence in diabetic mice’s glucose metabolism. Therefore, exercise therapy in the prevention and treatment of diabetes is effective and feasible.
