Ganoderma lucidum have significant biological diversity characteristics. It has important application value to study quality standards and fingerprints of Ganoderma lucidum from different varieties. Ganoderma lucidum in the north of Sichuan is a new Ganoderma lucidum (Leyss.ex Fr.) Karst.variety which were cultivated by sichuan agricultural university and Chengdu rongbao biological technology company, who used a wild ganoderma lucidum in the north of Sichuan. The ganoderma lucidum has been realizing large-scale cultivation for three consecutive years in Qingbaijiang district of Chengdu, and annual production is up to50tons. Based on the quality standard of ganoderma lucidum in "Chinese pharmacopoeia"2010edition, the paper researched systematically on the quality standard and fingerprints for ganoderma lucidum in the north of Sichuan.The first part of the paper, the ganoderma lucidum were identified by TLC, microscope, physicochemical method. And then the physical and chemical properties were discussed, the results showed that all the related items of test results are in line with the various index under ganoderma items in first2010version of "Chinese pharmacopoeia".The second part of the paper, the content of polysaccharide in ganoderma lucidum are determined. Determination method of Polysaccharide:the polysaccharide are detecded according to the determination method items of ganoderma polysaccha-ride in first2010version of "Chinese pharmacopoeia".and the methodology were reviewed.The results showed that RSD of the precision, reproducibility, stability were less than3.0%,and the recovery is good. The results showed that the content of the polysaccharide in ganoderma lucidum growing in the north of Sichuan are almost the same as the strains in the United States, Japan, South Korea, and the content all strains are greater than the prescribed limit (0.5%) in "Chinese pharmacopoeia"2010version.The third part of the paper, the content of ganoderma lucidum acid A were determined. Determination method of ganoderma lucidum acid A:A high phase liquid chromatography (HPLC) was used to determine ganoderma lucidum acid A. First the preparation method and HPLC analysis method of samples were determined, then the linear relationship has been inspected. The result showed that linear range of ganoderma lucidum acid A is in the range of0.010mg/mL-1.200mg/mL (r=0.9998), the RSD of methodological investigation results are less than3.0%, the average recovery is99.35%.The results showed that the content of ganoderma lucidum acid A in ganoderma lucidum in the north of Sichuan are higher than other strain, and the content of ganoderma lucidum acid A in ganoderma lucidum are different in different harvesting time and different recovery interval.The forth part of the paper, the content of four nucleoside were determined. Determination method of nucleoside:A high phase liquid chromatography (HPLC) was used to determine the nucleoside. First, the preparation method and HPLC analysis method of samples were determined, then the linear relationship has been inspected. The result showed that respectively, the linear range of Uridine, Inosine, Guanosine,Adenosine are0.0035~0.1400mg/mL,0.0160~0.0032mg/mL,0.007-0.1400mg/mL,0.0035~0.1400mg/mL; the average recovery of the four nucleoside are101.0%,101.0%,97.7%,100.0%; the RSD of methodological investigation results are less than3.0%.The results showed that there are Inosine in ganoderma lucidum in the north of Sichuan, and there are not in the other three fields, but the content of Uridine, Guanosine, Adenosine in the three strains are higher than in the north of Sichuan.The fifth part of the paper, the HPLC fingerprints of the liposoluble constituent in ganoderma lucidum were systematically studied. First, the preparation method and HPLC analysis method of samples were determined. Then the RSD of the method validation results for the precision, reproducibility, stability are less than3.0%. Finally, the HPLC fingerprints of the liposoluble constituent in ganoderma medicinal materials were established, by fingerprints analysis, the fingerprints of ganoderma lucidum in the north of Sichuan are very diffirernt from the other three strains. So, the common peak, a total of mode and evaluation method of liposoluble constituent were determined by comparing fingerprints of11groups of ganoderma lucidum in the north of Sichuan. The results showed that fingerprints similarity of liposoluble constituent of each batch samples were greater than0.900. Thus, the method in this paper is precise, stable, reproducible and easy to operate. It can be used for quality control and evaluation of Ganoderma lucidum.The sixth part of the paper, the quality standards (draft) of ganoderma lucidum in the north of Sichuan were initially formed. Compared With the quality standard, which included the character, identification, moisture, content of total ash, extract,the content of polysaccharide, for ganoderma lucidum in "Chinese pharmacopoeia"2010version, the ganoderma lucidum acid A, four nucleotide and the HPLC fingerprints of the liposoluble constituent were added to the quality standards (draft) of ganoderma lucidum.To sum up, the quality standards and HPLC fingerprints of ganoderma lucidum in the north of Sichuan were researched. The total contents of polysaccharide in the north of Sichuan are in accordance with the relevant requirement of "Chinese pharmacopoeia"2010version;The average contents of ganoderma lucidum acid A are in1.3697mg/g~5.5775mg/g range; The average contents of Uracil nucleoside are in0.1480mg/g~0.8819mg/g range; the average content of inosine are in0.0388mg/g~0.7118mg/g range; the average contents of guanosine are in0.1980mg/g~0.7491mg/g range, the average content of adenosine in0.0307mg/g~0.2290mg/g range; By the study of HPLC fingerprints of the liposoluble constituent,35common peak were estiblished, and the similarity of fingerprints for each batch of ganoderma lucidum herbs were greater than0.900. Therefore, the quality standard draft can basically reflect immanent quality of the ganoderma lucidum.The study can provide quality assurance for the cultivation, product development and application of ganoderma lucidum.
