Background:Cardiovascular diseases have become a very important one of thediseases that threaten human health, Coronary heart disease is the most hair acardiovascular disease,While atherosclerosis (journal Atherosclerosis, AS) is the basisfor the development of the lesions,In recent years, a large number of basic and clinicalresearch are support AS is an autoimmune disease and chronic inflammation of the newideas,That inflammation and immune responses to internal and external multiple riskfactors caused by AS onset, intermediaries and the central link in ruptured plaqueinstability. This view is based on the AS plaque inflammation,immune cells foundMMPs is the degradation of the ECM, leading to vascular middles mooth muscle cellinvasion and migration to the intima,and rarely express edin the normal coronaryvascular endothelial, but in a stable plaqueand unstable plaqueen do the lial expressionincreased,as well as by the regulation of TIMP-1andCD147, thus affecting thepathological process of the information of AS plaque.Objective:In this study, by immune his to chemistry chemical methods to detectMMP-9, TIMP-1and CD147expression in human normal coronary group andatherosclerosis in the coronary group, to observe the expression of MMP-9and TIMP-1,CD147expression and atherosclerosis relationship of the degree of progress.Provide theexperimental basis for the study of human coronary atherosclerosis pathogensis andtargets for the treatment.Methods:In accordance with the stenosis degree of60coronary artery specimensobtained from autopsy specimens, by visual Observation to determine the nine caseswith normal coronary arteries,51cases of atheroscler otic coronary artery, to isolate allcases and drawn, by HE staining and light microscope according to whether thelipidcore and itssizeis greater than40%of the patch area where the specimens weredivided into normal control group (9cases), stable plaque group (23cases), and unstable plaque group (27cases). Using chemistry to detect coronary artery vessel wallMMP-9and TIMP-1and CD147each expression.Results application SPSS13.0packagefor statistical analysis and processing, these threeindicators for monitoring using therank sum test, and pairwise comparison; correlation analysis using Pearson correlationanalysis.Result:The expression of MMP-9to express the positive parts of macrophages inatherosclerotic coronary intima, foam cells, fiber cell nucleus and perinuclearendothelial cell nucleus and nucleus, smooth muscle cells, medial smooth muscle cellsand fibroblasts.that only a very small amount of normal coronary endothelial expressionor no expression of the normal group and the stable group and the unstable group vary,but no significant differences between the stable group and the unstable group.TIMP-1in the normal coronary vascular smooth muscle expression. The positive parts of themembrane, AS in macrophages, fibroblasts and connective tissue cell expression, but itis a very small amount of expression. CD147expression on mono cytes, macrophagesand foam cells, some of the high expression.Normal coronary endothelial expression orthe expression of a very small amount. Pearson’s nonparametric rank correlation teststatistical analysis showed that MMP-9and TIMP-1, a positive correlation betweenCD147expression intensity of atherosclerotic coronary artery endothelial.Conclusion:1, in the normal coronary artery endothelial MMP-9, TIMP-1, CD147expression or a very small amount of expression; expression in coronary atherosclerosisand plaque instability group, the expression level was significantly higher than thestable group.MMP-9, TIMP-1, CD147expression reveals the three involved in thecoronary atherosclerotic plaque development, and MMP-9, CD147and plaqueinstability related.2, MMP-9in coronary atherosclerosis strong expression can promotethe development of atherosclerotic lesions,while MMP-9expression of CD147incoronary atherosclerotic plaques is positively correlated,indicating that CD147mayincrease MMP-9Expression.
