Objective: Use lentiviral RNA interference method to inhibit Oct4geneexpression in neural precursor cells derived from ES, in order to observe the function ofOct4gene in differentiation of Embryonic neural precursor cells to neurons in vitro.Method: First use of293T cell to get Oct4interference RNA interferencelentivirus slow virus, Second, use" five steps" method induce mouse embryonic stemcells differentiate to neural precursor,then give Oct4interference RNA lentivirus,using Real-time PCR method to Oct4expression of interferenced cell; Finally usingimmunohistochemical to count newborn neurons.Results: The experiments of Oct4interference lentiviral can effectivelytransferred into the neural precursor cells derived from ES, the expression of Oct4decreased by about40%, after induction of differentiation in normal group cells todifferentiate into neurons and about75.5+5.2%, GFP group differentiated neurons ofabout73.8+4.7%, Oct4interference of group newborn neurons are about33.8+2.9%.By SPSS10.0software for statistical analysis, blank control group compared with GFPgroup, P>0.05, not statistically significant; the blank control group compared to Oct4interference group, P <0.01, there was statistical significance; group GFP compared toand Oct4interference group, P <0.01, there was statistical significance.Conclusions: The use of lentivirus can transfer Oct4siRNA effectively into neuralprecursor cells, the Oct4expression in neural precursor derived from ES cells aresignificantly reduced. Confirmed in the ES source neural precursor cells still expressingOct4gene, Oct4gene is not only the maintenance of stem cell pluripotency but also anessential. factor in ES derived neural precursor cells differentiated into neurons.
