Effects Of Hypoxia On Invasion And Metastasis Of Hca-F Cells And Intervention Of Fucoidan

Objective: Tumor has been a serious hazard to human health. At present themain methods for tumor treatment are surgery, radiation and chemotherapy, but thesemethods have significant toxicity. Marine biological resource is rich and has themedicinal value. The development of Marine medicine has become the focus ofresearch and polysaccharides material of Marine creatures has many kinds of biologicalactivity.The purpose of this study was to determine effects of hypoxia on invasion andmetastasis of Hca-F cells and intervention of fucoidan.Methods:1.Chemical hypoxia model was established by Cobalt Chloride (CoCl2).The concentration of CoCl2were divided into five groups:50μ mol/L、100μ mol/L、150μ mol/L、200μ mol/L�'�400μ mol/L. MTT was used to screen treatmentconcentration and time of CoCl2.Hca-F cells were divided into three groups: control,hypoxia and hypoxia combined with fucoidan (500μg/ml) groups. The proliferation ofHca-F cells in different groups was evaluated by MTT assays.2. RT-PCR, Western Blotting and Immune fluorescence technique were used toexamine the expression level of protein and mRNA of hypoxia-inducible factor-1α(HIF-1α) and nuclear transcription factor-kappa B (NF-κB) in Hca-F cells. MTT andtranswell model were used to examine the adhesion, migration and invasion of Hca-Fcells and we should observe the change of cell behavior.Results: When the concentration of CoCl2was about100μ mol/L, the mRNA andprotein levels of HIF-1α and NF-κB were increased and the adhesion, migration andinvasion of Hca-F cells enhanced after24h. Fucoidan（500μg/ml）appeared to reduce theexpression of HIF-1α and NF-κB at both mRNA and protein levels on Hca-F cells underhypoxia. Moreover, fucoidan could lower the ability of adhesion, migration andinvasion of Hca-F cells. Conclusion: Hypoxia upregulates HIF-1α and NF-κB expression and promotesthe invasion and metastasis of Hca-F cells. Fucoidan have the effect of intervention.
