Objective:To culture human adipose mesenchymal stem cells and rabbit cornealendothelial cells in vitro, and to investigate the possibility of trans-differentiation ofhuman adipose mesenchymal stem cells to corneal endothelial-like cells throughco-culture the two kinds of cells in vitro, thus to find a new kind of seed for tissueengineering corneal endothelial cells reconstruction,and to lay a foundation for tissueengineering of corneal endothelial cells reconstruction.Methods:(1)The human adipose mesenchymal stem cells and rabbit cornealendothelial cells were cultured using inoculation or tissue culture methods, the twokinds cells were observed and identified using inverted microscope、flow cytometry、immunohistochemical methods.(2)Using transwell non-contract co-culture system toco-culture them for ten days.The expression of AQP1of the pre and post co-cultureendothelial cells were identified using immunohistochemical methods.Results:(1)Most of the cultured hADSCs were spindle, part of the region was acolony-like growth, showed a typical fibroblast-like.3rd generation hADSCs identifiedby flow cytometry results showed that CD44positive rate of93.7%and CD34positiverate of1.6%.(2)The cultured rabbit corneal endothelial cells were hexagon or polygon.The immunohistochemical staining showed that the nerve-specific enolase positiveexpression.(3)The inducted human adipose mesenchymal stem cells’ morphology didnot change significantly, and grew slowly. Immunohistochemical staining showed thatpositive expression of AQP1.Conclusion:(1)The human adipose mesenchymal stem cells and rabbit cornealendothelial cells can be cultured and passaged successfully in vitro.(2)The human adipose mesenchymal stem cells cultured in vitro can be induced intocorneal endothelial-like cells.
