Objective: Multiple organ dysfunction syndrome(MODS) is a kind of clinicalsyndrome causes dysfunction within2or more than2organs simultaneously or insequece after the body suffer from traumas, shock, serious infection, or get large-scaleoperations in24hours within more than2organs. It is one of the main reasons that leaddying patients to death. MODS becomes an international research target on account ofits instant occurrence, high incidence and deadly results. This experiment was set on thebasis of one of “Four Method”, that is——Fuzheng Guben principle to observe theinfluence of Shenqi Fuzheng Injection, which represents the Fuzheng Guben Chinesemedicine, on CEC and TGF-β1in the MODS model of rats. Moreover, this paper willdiscuss the protective effect of Shenqi Fuzheng Injection on viscera of rats through boththe vibrating indexes of them: ALT, CR and the pathological comprehensive conclusionof lung, liver and kidney. In this way, the new experimental foundation should beprovided to clinical treatment on MODS.Methods: Grouping96male SD rats into3units at random as sham operationgroup (group J,32rats), MODS group (group M,32rats), and treatment group (group S,32rats, using Shenqi Fuzheng injection). Furthermore, every group was divided as4subgroups at random in according to four time indexes:12hours,24hours,36hours,and48hours, with8rats in every group. The rat MODS model was created by means ofischemia-reperfusion combined with zymosan intraperitoneal injection: firstly clampingthe superior mesenteric artery with artery clip for45minutes, and then opening theartery to restore perfusion, and12hours later injecting small doses of zymosan(125mg/kg). At the end of experiment, SPSS17.0would be utilized to analyze allstatistics. After being injected small doze of zymosan respectively on time point of0.5-hour,12-hour,24-hour and36-hour, the treatment group would take2ml/200g of Shenqi Fuzheng injection, and the MODS group2ml/200g of normal saline. And afterthe membrane was formed respectively on time point of12-hour,24-hour,36-hour and48-hour, a particular number of rats would be killed to take their blood sample. With thesample, TGF-β1would be inspected by the method of double antibody ABC-ELISA,and CEC by Flow Cytometer at the same time being marked fluorescence antibody. It isthe same with ALT, CR. The tissue specimen of liver, lung, kidney of rats from eachgroup48h were taken to make relevant histopathologic examination.Results:1. After MODS model was constructed, rats from group M and group Sbecame upset, anorectic, irritated to some extent, and got sick with bouffant dull fur,twisting body, weariness, and tachypnea, although in different times. However, ratsfrom group J had no or only had such reactions slightly.2. Variable CEC amounts inevery group were distinct from each other and thus were proper to adopt as statistics:distinctions between group12h-J and group M, group M and group S had statisticalsignificance (P <0.05). And so it is with distinctions between group24h-J and group M,group M and group S; group36h-J and group M, group M and group S (P <0.05); group48h-M and group J, group S and group M, group S and group J (P <0.05).3. VariableTGF-β amounts in every group were distinct from each other and thus were proper toadopt as statistics: distinctions between group12h-J and group M, group J and group Shad statistical significance (P <0.05). And so it is with distinctions between group Mand group J, group S and group J, group S and group M (P <0.05) respectively on timepoint of24hours,36hours and48hours.4. The index of ALT, CR on every phasepoint of group M and group S was higher than that of group J, which means a statisticalsignificance (P <0.05).5. Pathological sections of organs of rats from group48h-S showfaster recovery than that from group48h-M.Conclusions:1. MODS models of rats were successfully created: the amount ofCEC contained in plasma obviously increased when rats got MODS, which figured outthat chrotoplast in blood vessel were damaged.2. This experiment was set on the basisof two of “Four Method”, that is, acute deficient syndrome and Fuzheng Gubenprinciple to observe the preliminary influence of Shenqi Fuzheng injection, whichrepresents the Fuzheng Guben Chinese medicine, on CEC and TGF-β1in the MODSmodel of rats.3.Shenqi Fuzheng injection, a representive of Fuzheng Guben Chinesemedicine could effectively lower the amount of CEC in rats from treatment group,which showed that the injection could protect chrotoplast in blood vessel from beingdamaged.4. The existing expression of TGF-β1during early phase increased when rats got MODS. However, Fuzheng Guben Chinese medicine had effect on adjustingcontainment of TGF-β1thus influencing immune system of rats to regulate theirinflammatory response.5. Shenqi Fuzheng injection, a representive of Fuzheng GubenChinese medicine could protect viscera organ function of rats with MODS from furtherdeterioration.
