Objective:The ovarian tumor to be one of the three most malignant tumor infeminine reproductive system, because it early clinical feature is not obvious as well asthe progress is quick, the ovarian tumor already became the main murderer whichendangers the feminine life. Now, the main treatment of ovarian tumor is surgery andchemotherapy, but the prognosis of patients is still not ideal. Therefore, seeks the newmethod of treatment to be imperative. The estrogen as the ovarian main steroid productof, have a close relationship with the high risk of ovarian cancer. Therefore, understandthe mechanism of between estrogen and ovarian tumor as well as the anti-estrogeneffect, is most important to the explore a new therapeutic schedule of ovarian tumor.This research is for the purpose of discussing the estradiol and estrogen inhibitorICI182,780to the SKOV3cell multiplication, perishes weakly as well as the proteinexpression influenceThe main purpose of this study is to investigate the effects of estradiol and anti-estrogen ICI182,780on the proliferation, apoptosis and protein expression in humanepithelial ovarian cancer SKOV3cells.Methods:After interfered by E2and E2+ICI182,780, apoptosis of SKOV3cellwas detected by Annexin V/PI. Protein expression of SKOV3cell were analyzed byWestern-Blot; observe the protein expression of SKOV3cell through immunityfluorescence microscope.Results:E2inhibited the apoptosis of SKOV3cell after24and48h cultured; E2effect the expression of ERα and protein on the PI3K/AKT signal pathway, anti-estrogen ICI182,780counteract the effect of E2on the SKOV3cell.Conclusion:E2could inhibit the apoptosis of SKOV3cell. Raise the proteinexpression of p-AKT, p-PTEN,p-GSK3β, cyclinD1; reduce the protein expression ofERα; anti-estrogen ICI182,780counteract the effect of E2on SKOV3cell, both of them have no influence on the expression of AKT and GSK3β.
