【Objective】To observe the expression&significance of autophage&ER stressmarkers on the hepatic injuries model of mice induced by LPS and GalN and toinvestigate its function in the hepatic injury and the relativity of autophage&ERstress.【Methods】34male Kunming mice in clear grade with similar weight (25g±5g),were divided randomly into two groups:(1) normal control group (n=8):0.9%NaCl0.2ml each i.p, angular blood from the left eyes and take out of the liver;(2) hepatic injury model group (n=26),(LPS5ug+GalN0.02g) each i.p, and afterinjected LPS/GalN1h,3h, and6h, extract the latter group angular blood from the lefteyes and inject into centrifuge tube, take out of the liver and freeze in liquid nitrogentanks in anesthesia with Pentobarbital Sodium.Plasma ALT activity was detected by Reitman method. The expression of eachgroup’s liver LC3、Beclin-1and CHOP was detected by Western-blot.【Results】The content of the plasma ALT in the1h&3h group of hepatic injurygroup and normal control group have no significant differences (P <0.05); Theplasma ALT activity of the6h group in hepatic injury group was much higher thanthat of the normal control group(P <0.001). The expression of mouse liver tissue’sLC3II、Beclin-1: the expression of LC3II、Beclin-1enhance significantly, peak in3h,attenuate in6h after stimulus with LPS/GalN. The expression of mouse liver tissueCHOP: the expression of CHOP enhances significantly, peaks in3h, dropssignificantly in6h after stimulus with LPS/GalN.【Conclusions】(1) The expression of autophage in hepatic injuries rise, but the expression will drop at the later period of liver injuries if there were critical hepatosisand hepatic failure. The autophage has protective effect in hepatic injuries, thedropping in the later period possibly has relationship with hepatic energy depletion.(2) The expression of chop in hepatic injury induced with LPS/GalN rises, but theexpression drops significantly in the later period of hepatic injuries if there if therewere critical hepatosis and hepatic failure. The ER stress markers have protectiveeffect in hepatic injuries, the dropping in the later period possibly has relationshipwith critical liver cell death, then cell organelles death including endoplasmicreticulum.
