ObjectiveTo study the influence of hyaluronidase (HYD) activity and CRISP2gene mRNA expression level in SD rats mature sperm after different dose X-ray irradiation in different time points and compare relationship with sperm motility。 And discuss the effects of sperm motility mechanism, further provide experiment basis of study the effect of male reproductive function after ionizing radiation.MethodRandomly divided into five groups and each big group by1、4、8、12、24h time point and divided into five groups.with0、1、2、4、6Gy.With x-ray local exposure SD rats the reproductive system (dose rate:442.89cGy/min) from Precise linear accelerator, we respectively used modified gelatin substrate membrane method and PCR technology to test hyaluronidase activity and CRISP2gene mRNA expression level in rats mature sperm,detect rats mature sperm motility with conventional microscope.Result1. HYD activity rats mature sperm after the X-rays.HYD positive reaction rate in each time point in control group was (70.2±1.92)%、(67.6±5.81)%、(69.8±2.16)、(68±6.67)%、(71.2±2.86)%, that of4、6Gy x-ray group in different time (1、4、8、12、24h) dramaticlly falled than control group (6.28<t<25.8, P<0.05), HYD reaction rate is lowest after24h in6Gy group minimum (t=25.8, P<0.01); the difference of HYD positive reaction rate was not statistically significant (0.47<t<=1.313, P>0.05) in2Gy X-ray irradiation in different time (1、4、8、12、24hours); HYD positive reaction rate of the difference time was not statistically significant (P>0.05) in every exposure group。2. CRISP2gene mRNA expression level after the X-rays. The level of rat sperm CRISP2mRNA exposed4、6Gy x-rays group was significantly lower than the control group(P<0.05); including6Gy exposure24hours after relative express quantity minimum (P<0.01); When accept2Gy after irradiation,the level of rats CRISP2mRNA have reduced in different degree, but significant changes occur in exposure after8h (P<0.05); in lower dose(1Gy) CRISP2mRNA were not changed significantly (P>0.05).3. The relationship between sperm vitality and CRISP2gene mRNA and the HYD after the X-rays.The sperm motility (A±B%) is78.37±4.046%in the control group(0Gy exposure),1、2Gy and4Gy(1、4、8h)exposure sperm motility group did not decrease significantly (P>0.05);4Gy(12、24h)and6Gy exposure group sperm motility is obviously lower than those of control (P<0.05); Sperm vitality and CRISP2gene mRNA or HYD concluded in a straight line by linear related statistical analysis, testing the line, the correlation coefficient was respectively R=0.802(0.944)、F=41.37(150.47),(P<0.05)Conclusion1. Sperm vitality in SD rat after different doses x-ray exposure can result decline, which likely related with CRISP2gene mRNA related expression levels and HYD activity.2. Experiment examination of sperm motility might be more sensitive than pure sperm routine experiment parameter.3. Sperm CRISP2relative gene mRNA expression after certain dose range X-ray(2gy) irradiation was different decline degree in different time points, this is related due to likely ultimate effect of the ionizing radiation damage.
