The Research Of Expression And Clinical Significance Of Cystatin SN、MMP-11in Gastric Cancer

Objective:This study aimed to investigate the expression of Cystatin SN and MMP-11in gastric cancer tissues, and explore their correlation with the degree of malignancy in gastric cancer.Methods:The Cystatin SN and MMP-11mRNA expression was examined by using real-time quantitative polymerase chain reaction on a cohort of human gastric cancer tissues (n=40) and corresponding adjacent tissues (n=40). Relationship of their expression with patients’age, gender, tumor site,tumor size, clinical stage,the degree of malignancy, degree of infiltration and lymph node metastasis was analyzed. The correlation between cystatin SN and MMP-11was also analyzed.Results:1. The expression level of Cystatin SN mRNA in gastric cancer was dramati-cally increased compared with the corresponding adjacent tissues (P<0.05);The expression of Cystatin SN was significantly associated with tumor size,lymphnode metastasis,depth of invasion and gastric cancer stage(P<0.05); may have a bit relation to degree of tumor differentiation(p=0.057).However, the expression of Cystatin SN had no correlation with the age and gender of patients in this study(P>0.05).2. The expression level of MMP-11mRNA in gastric cancer was dramatically increased compared with the corresponding adjacent tissues (P<0.05);The expression of MMP-11was significantly associated with lymphnode metastasis,depth of invasion and gastric cancer stage(P<0.05);However the expression of MMP-11had no correlation with size and differentiation of tumor, age and gender of patients in this study(P>0.05).3. Correlation analysis showed that the expression of Cystatin SN and MMP-11in gastric cancer had a positive correlation(r=0.449, P<0.05).Conclusions:1. The expression level of Cystatin SN and MMP-11mRNA in gastric cancer was dramatically increased compared with the corresponding adjacent tissues, and they were significantly correlated with the development of gastric cancer.2. The overexpression of Cystatin SN and MMP-11mRNA had a close relationship with depth of invasion, lymphnode metastasis and tumor stage of gastric cancer, which may participate in pathogenesis of invasion and metastasis of gastric cancer.3. Cystatin SN and MMP-11had an interoperability in the progress of gastric cancer, joint detection of Cystatin SN and MMP-11may become an important molecular maker to judge the recurrence and metastasisdegree of gastric cancer. At the same time, it will provide a new thinking for biological targeted therapy of gastric cancer.
