Objective To observe the expression of netrin-1protein in retina of STZ induceddiabetic mice,evaluate the relativity between netrin-1and the early stage of diabeticretinopathy.Methods To select60male C57BL/6mice of good health,distinguish them to be3group,45in diabetic group and15in control group.Establish STZ-induced diabeticmice model by intraperitoneal injection of streptozocin(STZ);Measure the bloodsugar and weight in different stage of diabetes,HE stain will be used to observe thepathology change of early diabetic retinopathy,use immunofluorescence to observethe expression of netrin-1protein in different times of diabetic retinopathy.Results The blood sugar of diabetic group is much higher than control in4,8and12weeks after injection;in diabetic group,the blood capillary vessel wall isthick,limiting layer get to be thick,ganglion cell engorge and disorder,and there ismicroangioma growing to cavum vitreum.4,8and12weeks after injection.Theexpression of netrin-1protein is mainly expressed in GCL and INL.There is noexpression of netrin-1protein in WESTERN-BLOT.Conclusion STZ-induced diabetic mice model is suitable for the study of earlydiabetic retinopathy.The pathology change of retina is manifest,and there ismicroangioma.The expression of netrin-1protein in diabetic group is higher thancontrol group,which is the same as the expression of Netrin-1mRNA. Regulation ofnetrin-1in angiogenesis and axon guidance offers new hope in pathogenesy researchand clinical treatment of correlated disease.
