Objective To investigate the expression level and clinical significance of the candidatetumor suppressor gene OPCML in human breast cancer tissues and cell lines (MDA-MB-231,MB468,MCF-7,SK-BR-3,T47D,BT-549).Methods1.With tissue microarray technology and PV immunohistochemistry,theexpression of OPCML was evaluated in129cases of breast cancer,41cases of adjacent tissueand49cases of benign breast diseases tissue,and analyze the relationship between clinicalopothological features, general regulation index of breast cancer and the expression ofOPCML.2.Western blot was applied to evaluate the expression of OPCML in six cases ofbreast cancer cell lines (MDA-MB-231, MB468, MCF-7, SK-BR-3, T47D, BT-549).Results1. Compared with the benign breast diseases tissue and tumor adjacent tissue,the positive rate of OPCML in the breast cancer was significantly lower (75.51%,90.24%vs20.16%, P<0.05).2.The expressions of OPCML was positively related with histological grade,pTNM staging and lymphoid node metastasis of breast carcinoma (P<0.05). Significantlynegative correlation was found between the expressions of OPCML andKi67(r=-0.236,P<0.05)、HER2(r=-0.262,P<0.05)、EGFR(r=-0.243,P<0.05).3.Western blottingresults showed that OPCML loss expression in the six kinds of breast cancer celllines(MB231,MB468,MCF-7,SK-BR-3, T47D,BT-549), except the adjacent tissues.Conclusion1.OPCML protein is highly expressed in benign breast lesions and tumoradjacent tissue with low expression in breast cancer. The expression of OPCML protein、Ki67、 HER2and EGFR were negatively correlated in breast cancer.OPCML proteinexpression level was positively correlated with the histological grade, pTNM staging is significant and lymphoid node metastasis of breast cancer.2.Western blotting results showedthat OPCML loss expression in the six kinds of breast cancer cell lines(MB231,MB468,MCF-7,SK-BR-3, T47D,BT-549), except the adjacent tissues. In summary, OPCML is atumor suppressor gene in breast cancer and closely associated with the carcinogenesis anddevelopment of breast carcinoma,and could be an independent prognostic indicator.
